链霉菌H8797产生的二酮哌嗪类化合物的分离纯化和结构鉴定

摘  要:目的研究链霉菌H8797发酵液中抑制LFA-1/ICAM-1介导的人B淋巴瘤JY细胞聚集的活性成分。方法利用HP-20大孔吸附树脂吸附发酵液中的活性成分,梯度解吸,分部收集,跟踪监测活性,再通过C18硅胶柱层析和LH-20柱层析分离,高效制备液相色谱精制,测定活性,通过紫外、红外、元素分析、高分辨质谱、核磁共振进行分析,得到活性单组份。结果分离纯化了活性化合物H8797A并对其进行了结构确证。结论化合物H8797A为首次从天然产物中分离获得,属于二酮哌嗪类化合物,具有抑制LFA-1/ICAM-1介导的人B淋巴瘤JY细胞聚集的活性。Objective To study the active compound which inhibits the LFA-1/ICAM-1 mediated human B lymphoma JY cell aggregation from fermentation broth of Streptomyces sp. H8797. Methods Active components were absorbed by macroporous absorption resin HP-20 and gradient eluted, fraction collection, tracking and monitoring activity. Purified component were isolated by LH-20 column chromatography and preparative HPLC. Active compound was elucidated by elemental analysis and spectral analysis. Results The active compound H8797A was isolated and identificated. Conclusion H8797A was identified as a diketoperazine derivative and isolated from natural products for the first time. It inhibits LFA-1/ICAM-1 mediated human B lymphoma JY cell aggregation.
