产黄青霉原生质体融合及融合子初步分析

摘  要:目的以青霉素工业生产菌株产黄青霉为研究对象,对丝状真菌原生质体融合及其融合子进行了初步的研究。方法通过紫外线诱变和基因转化筛选出具有不同遗传标记的2株亲本菌株,对供体亲本菌株原生质体热灭活后进行原生质体融合,并对融合子进行了分离及传代分离。结果在选择性培养基上筛选到5株融合子,融合率为2.97×10-5,融合子分离和传代分离后产生亲本菌株分离子和异核体。结论 5株融合子均为异核体,在自然条件下融合子很难形成杂合二倍体和重组子。Objective Filamentous fungi protoplast fusion Penicillium chrysogenum of penicillin industrial strain. Methods and the fusants were preliminary studied for the Parental strain I were selected from original strains radiated by ultraviolet rays, and parental strain II was abtained by transformation of original strains with amdS gene. 2 parental strains got different genetic marks. Protoplasts fusion using polyethylene glycol was carried out with parental strain II protoplasts inactivated by heated, and the fusant strains were achieved by plating fusion protoplasts on the selection medium in which the sole nitrogen source was acetamide. The fusants were segregated and generated. Results 5 fusants regenerated in the selective medium. The fusion rate was 2.97~ 10-5. The segregants were parental type strains and heterokaryons by the phenotypic or genotypic characterization. Conclusion 5 fusants were heterokaryons, and it was difficult that fusants formed heterozygote and recombinant haploid when generated under natural condition.
