摘 要:目的 研究玉溪市2010-2012年乙型流感流行病毒株的抗原性和基因特点。方法 通过MDCK细胞分离培养乙型流感病毒,血凝抑制试验检测其抗原性,RT-PCR克隆HA1基因,测序后通过软件MEG5.0进行基因进化研究,同时与WHO推荐的乙型流感病毒疫苗株进行比较。结果 玉溪市乙型流感病毒的流行2010年以BV谱系为主,2011年BV和BY谱系同时流行,2012年以BY谱系为主。基因分析:BY系毒株与B/Wisconsin/01/2010的碱基同源性在99.5%~99.7%之间;BV系毒株与B/Brisbane/60/2008碱基同源性在98.7%~99.1%之间。与疫苗株比较,流行株发生了氨基酸替换。结论 建议2012年玉溪市使用WHO推荐疫苗株B/Wisconsin/01/2010进行乙型流感病毒流行的防控和储备。Objective The aim of this research was to study the genetics and antigenic characteristics of the influenza B virus in Yuxi from 2010 to 2012. Methods The influenza B virus was isolated and cultured using MDCK cells, the antigenicity was analyzed by the hemagglutination inhibition (HI) test, the phylogenetic tree of HA1 gene was constructed using software MEG5.0 after se- quencing the RT-PCR products, and the influenza B virus vaccine strains was compared with WHO recommendation. Results For the epidemic of influenza B virus, the BV lineages were epidemic on 2010, both the BY and BV lineages were epidemic on 2011, and the BY lineages were epidemic on 2012. For the genetic background, the nucleotide homologies between BY virus strains and the B/Wisconsin/01/2010 strain were from 99.5% to 99.7%, the nucleotide homologies between BV virus strains and the B/Bris- bane/60/2008 strain were from 98.7% to 99.1%. Compared with the vaccination strains, there were amino acid substitutions in epidemic strains. Collclusion The vaccination strain B/Wisconsin/O1/2010 recommended by WHO needs to be reserved in order to prevent and control the influenza B virus in Yuxi starting from 2012.
