摘  要:目的对改良TSA-L培养基进行改进,建立一种配方简单、成本低廉及适合结核分枝杆菌L型(Mycobacterium Tuberculosis L forms,MTB-L)快速生长的培养方法。方法在改良TSA-L培养基中加入最适比例的有利于分枝杆菌生长的卵磷脂(Lecithin)和胆固醇(Cholesterin),制备LC-MTB-L培养基;采用改良TSA-L培养基和LC-MTB-L培养基接种培养肺科门诊患者痰标本,比较两种培养基的MTB-L阳性检出率和培养阳性所需时间。结果 LC-MTB-L培养基中卵磷脂和胆固醇的最适加入量均为0.6 g;LC-MTB-L培养基MTB-L阳性检出率显著高于改良TAS-L培养基(P<0.05),同时其培养阳性所需时间显著低于改良TAS-L培养基(P<0.05)。结论 LC-MTB-L培养基配方简单、成本低廉、MTB-L生长快速,适合临床推广应用。Objective The object of this study was to improve the modified TSA-L medium, in order to establish a novel medium suitable for rapidgrowing of Mycobacterium tuberculosis L forms (MTB-L) with simple composition and low cost. Methods LC-MTB-L medium was prepared by adding optimal proportion of lecithin and cholesterin into the modified TSA-L medium. Sputum from tuberculosis patients were inoculated into modified TSA-L medium and LC-MTB-L medium respectively. Positive detection rates of MTB-L and duration for positive culture in the two media were analyzed. Results Optimal proportion of lecithin and cholesterin was 0.6 gram eachy in LC-MTB-L medium. Positive detection rates of MTB-L in LC-MTB-L medium were higher than those in modified TAS-L medium (P〈0.05). Meanwhile, durations for positive culture of MTB-L in LC-MTB-L medium were shorter than those in modified TAS-L medium (P〈0.05). Conclusion LC-MTB-L medium is suitable for MTB-L rapid growth with simple composition and low cost, which is worthy to be popularized and applied in clinics.
