摘 要:为草科鸡的品种选育及品种改良提供理论依据,以MSTN基因作为候选基因,运用PCR-SSCP和DNA测序技术对MSTN基因进行扫描,研究其多态性位点与生长性状之间的关联性。结果表明:外显子1上有2 100bp(C/T)和2 109bp(C/T)2个突变位点,但未引起氨基酸变异,为同义突变;内含子1上有4 295bp(C/T)、4 359bp(C/T)、4 404bp(A/C)3个突变位点。外显子1表现出CC、CT、TT(2 109bp)和CT、TT(2 100bp)5种基因型,不同基因型间生长性状并没有表现出显著性差异;内含子1表现出CT(4 295bp)、CT(4 359bp)和CC、CT、TT(4 404bp)5种基因型,4 295bp和4 404bp位点TT基因型个体的胸肌率均显著高于CT基因型个体(P<0.05),4 359bp位点TT基因型个体的半净膛率显著高于CT基因型个体(P<0.05)。外显子1上有CC、CT、TC和TT共4种单倍型,内含子1上有CCC、CCT、CTC、CTT、TCC、TCT、TTC和TTT共8种单倍型。MSTN基因可作为影响草科鸡生长性状的候选基因。To provide a theoretical basis for variety breeding and improving the variety of Caoke chicken,the research was carried out by using the MSTN gene as a candidate gene,and subsequently scanning the myostatin (MSTN ) gene with PCR-SSCP and DNA sequencing techniques in order to investigate the influence of MSTN gene on growth traits of Caoke chicken.The results showed that there were two loci (C2100T and C2109T)in exon 1 showing five genotypes CC,CT,TT(C2109T)and CT, TT(C2100T).There were three loci (C4295T,C4359T and A4404C)in introns 1 showing CT(4 295 bp), CT(4 359 bp),CC,CT and TT(4 404 bp)five genotypes.The two mutations in exon 1 as synonymous mutations did not cause the amino acid variations,which had no effections in growth traits of Caoke chicken.While the three loci in introns 1 had effects on the rate of chest muscle and the chamber half net rate in chicken.Four haplotype (CC,CT,TC,TT)in exon 1 and eight haplotype (CCC,CCT,CTC, CTT,TCC,TCT,TTC,TTT)in introns 1 of Caoke chicken were found.All the results implied the MSTN gene could be a candidate gene.
