摘  要:目的观察慢性间歇低氧对大鼠血脂及血管内皮和下丘脑ghrelin表达的影响。方法将健康雄性Wistar大鼠60只,随机分为正常空气组,持续低氧组和间歇低氧组,每组20只,第1、3个月末测大鼠血脂,C反应蛋白(CRP),RT-PCR法测下丘脑和血管内皮ghrelin基因表达,Western blot法测血管内皮血管细胞黏附分子1(VCAM-1)、细胞间黏附分子1(ICAM-1)蛋白表达。结果与正常空气组比较,持续低氧组血脂、CRP无显著变化(P>0.05),间歇低氧组血脂、CRP明显增加(P<0.05);与正常空气组比较,持续低氧组下丘脑和血管内皮ghrelin基因表达无显著差异(0.69±0.35 vs 0.68±0.41,1.28±0.32 vs 1.19±0.21,P>0.05),间歇低氧组下丘脑和血管内皮ghrelin基因表达明显降低(0.52±0.28 vs 0.68±0.41,0.69±0.21 vs 1.19±0.21,P<0.05);与正常空气组比较,持续低氧组VCAM-1、ICAM-1蛋白表达无明显变化(P>0.05),间歇低氧组VCAM-1、ICAM-1蛋白表达明显增加(P<0.05)。结论间歇低氧作为应激源,可能经ghrelin-血脂、炎症途径参与AS的发生、发展进程。Objective To study the effect of chronic intermittent hypoxemia on lipid level and ghrelin expression in endothelium and hypothalamus of rats.Methods Sixty healthy male Wistar rats were randomly divided into normal group,persistent hypoxemia group,and intermittent hypoxemia group.Their body weight,serum lipid and CRP level were measured,expressions of ghrelin,VCAM-1 and ICAM-1 protein in hypothalamus and endothelium were detected by RT-PCR and Western blot,respectively,at the end of months 1 and 3.Results No significant difference was found in body weight,serum lipid and CRP level between normal group and persistent hypoxemia group(P>0.05).The body weight,serum lipid and CRP level were significantly higher in intermittent hypoxemia group than in normal group(P<0.05).No significant difference was found in ghrelin expression level between normal group and persistent hypoxemia group (P > 0.05).The ghrelin expression level was significantly lower in intermittent hypoxemia group than in normal group(P<0.05).No significant difference was found in VCAM-1 and ICAM-1 expression level between persistent hypoxemia group and normal group(P>0.05).The VCAM-1 and ICAM-1 expression levels were significantly higher in intermittent hypoxemia group than in normal group(P< 0.05).Conclusion Decreased ghrelin expression in endothelium is related with increased expression of inflammatory factors.As a stress source,intermittent hypoxemia participates in the occurrence and progress of atherosclerosis through the ghrelin/lipid and inflammation pathways.
