摘  要:目的探讨吡那地尔预处理对大鼠皮肤-肌肉切口牵拉(SMIR)术后周围组织神经生长因子(NGF)和葡萄糖转运蛋白1(GLUT-1)表达的影响。方法成年雄性SD大鼠24只,随机均分成对照组、假手术组、SMIR组和吡那地尔25μg/kg+SMIR组(吡那地尔组)。假手术组只切割皮肤、肌肉,SMIR组在假手术组基础上牵拉皮肤、肌肉1h,吡那地尔组腹腔注射吡那地尔后1h制成SMIR模型。Western blot检测各组切口周围组织NGF和GLUT-1的蛋白表达。结果与对照组比较,SMIR组NGF表达增加,GLUT-1表达降低(P<0.01);假手术组GLUT-1表达降低(P<0.05)。与SMIR组比较,吡那地尔组NGF表达降低,GLUT-1表达增加(P<0.05)。结论吡那地尔预处理改善SMIR术后切口周围组织局部微环境,可能与NGF和GLUT-1表达变化有关。Objective To investigate the effects of pinacidil preinjection on tissue nerve growth factor(NGF)and GLUT-1expressions after skin-muscle incision and retraction(SMIR)in rats.Methods Forty male rats were equally randomized into 4groups of A(blank control),B(sham operated),C(SMIR for 1hour)and D(pinacidil 25μg/kg preinjected before SMIR).The protein expressions of NGF and glucose transporter-1(GLUT-1)in the tissues around the incision were detected by Western blot.Results The protein expression of NGF was higher,but that of GLUT-1was lower,in group C than those in group A(P〈0.01).The protein expression of GLUT-1was lower in group B than that in group A(P〈0.05).The protein expression of NGF was lower,but that of GLUT-1was higher,in group D than those in group C(P〈0.05).Conclusion Preinjection of pinacidil 25μg/kg may improve the local microenvironment around incision after SMIR,which may be related to the changes in the expressions of NGF and GLUT-1in the tissues around the incision in rats.
