摘 要:目的探讨甘油磷酸氧化酶—过氧化物酶(GPO-PAP)法测定冰冻解冻去甘油红细胞甘油残留量的可行性。方法利用回收试验、重复性试验、干扰试验、线性试验对该方法学进行评价,并应用该方法对本血站的冰冻红细胞产品进行甘油残留量抽检验证。结果回收试验平均回收率为99.7%;重复性试验批内CV<5%;干扰试验显示Hb≤10.0 g/L,对本方法结果无干扰;线性试验表明有较好的线性关系(r=0.9998);该方法对本血站的冰冻红细胞产品的甘油残留量抽检结果符合国家质量标准的要求。结论该方法准确快捷,容易操作,解决了传统方法测定甘油残留量的不足,值得推广。Objective To evaluate the GPO-PAP method and its feasibility for measuring the glycerol residue in deglycerolized red blood cells (RBCs). Methods The recovery testing, repetitive testing, interference testing, and linear testing were conducted to evaluate the GPO-PAP method. Besides, the GPO-PAP method was used to detect the glycerol residue in deglyeerolized RBCs. Results The average recovery rate was 99.7% ;repeat tests was within CV〈5%; interference test showed that Hb ≤ 10.0 g/L had no interference on the measurement;linear test showed a good linear relationship(r=0.9998). This method was applied to measure the glycerol residue on the frozen blood erythrocytes and the results were in line with national quality standards, stable and accurate. Conclusion The GPO-PAP method is a fast, accurate, and easily to handle method to detect the glycerol residual, and can overcome the shortcoming of the traditional method and is worth to promote.
