摘  要:目的探讨子宫内膜异位症与健康对照组之间的血清差异表达蛋白,为子宫内膜异位症早期诊断提供生物标志物的检测方法。方法选择临床明确诊断的子宫内膜异位症患者以及健康妇女(对照组)各20例,采集其血清利用同位素标记相对和绝对定量(iTRAQ)技术检测其血清差异蛋白,并进行功能注释及生物信息分析。并从中选择明显相关的蛋白,利用Western-blot进行验证。结果与对照组相比,子宫内膜异位症患者血清中有29个蛋白异常表达,其中4个蛋白表达明显降低,25个蛋白表达明显升高。Western-blot验证结果与iTRAQ结果相符。结论证实了iTRAQ技术对于筛选生物标记物具有实用性。CD44,纤维蛋白原-α和纤连蛋白1等参与细胞粘附的分子在子宫内膜异位症的发病机制中起作用。Objective To identify differentially expressed proteins in the serum of women with and without endometriosis and determine the pathogenesis of endometriosis. Methods The serum samples from 20 patients with endometriosis and from 20 healthy women without endometriosis were analyzed using iTRAQ technology in order to detect the differential expression of proteins. The validation of the proteins was performed using Western blot. Results Compared with the control group, 29 proteins were abnormally expressed in endometriosis, of which 4 proteins were down-regulated and 25 proteins were significantly up-regulated. The relative increases in the serum protein levels of five of the identified proteins were validated by Western blot. Conclusion Most of these proteins differentially expressed which is consistent to the results from previous studies. The results of this study has demonstrated the utility of the iTRAQ technique for biomarker discovery. The proteins CD44, fibrinogen-αand fibronectin-1 participate in the biological process of cell adhesion, which could have resulted in the malignant behaviors of endometriosis.
