摘  要:目的探讨Cr(Ⅵ)染毒对A549细胞中hMSH2和p53mRNA表达的影响。方法体外培养A549细胞,实验组用脱氢抗坏血酸孵育90min,对照组常规培养,然后两组均用浓度分别为0.00、1.25、2.50、5.00μmol/L的Cr(Ⅵ)染毒。用实时聚合酶链反应(RT-PCR)检测A549细胞中hMSH2和p53mRNA的表达量。结果实验组与hMSH2和p53mRNA的相对表达量均低于对照组,差异均有统计学意义(P<0.05);Cr(Ⅵ)剂量依赖性地降低细胞中hMSH2和p53mRNA的表达;脱氢抗坏血酸预处理可以进一步降低暴露于Cr(Ⅵ)的A549细胞中hMSH2mRNA的表达。结论脱氢抗坏血酸在Cr(Ⅵ)致细胞毒性中起促进作用;Cr(Ⅵ)剂量依赖性地降低细胞中hMSH2和p53mRNA的表达,因此Cr(Ⅵ)可能参与肺癌细胞癌变过程。Objective To explore the influence of Cr(Ⅵ) exposure on expression of hMSH2 and p53 mR- NA of A549 cells. Methods A549 cells were cultured in vitro, the cells in experimental group were incubated with dehydroascorbic acid for 90 min, and cells in the control group were cultured by conventional meth- od. The cells were exposed to 0. 00, 1.25, 2.50 and 5. 00 μmol/L Cr(Td) respectively, and the expression of hMSH2 and p53 mRNA in A549 cells were detected by Real--Time PCR. Results The relative expression of hMSH2 and p53 mRNA in the experimental group were lower than that in the control group, and the differ- ence was statistically significant(P〈0.05). The expression of hMSH2 and p53 mRNA in A549 cells decreased with Cr(VI) exposure in a dose--dependent manner The expression of hMSH2 in A549 cells exposed to Cr (1/I) was further reduced by pretreatment with dehydroascorbic acid. Conclusions The dehydroascorbic acid plays a catalytic role in the cytotoxicity induced by Cr( Ⅵ ) ; The expression of hMSH2 and p53 mRNA in A549 cells decreased with Cr(Ⅵ) exposure in a dose--dependent manner, thus Cr(Ⅵ) may participate in the carcinogenesis process of lung cancer.
