草麻黄抗补体组份对血小板变形及内皮细胞损伤作用

摘  要:目的从草麻黄中分离抗补体组份并观察其抗补体依赖性的损伤作用。方法采用柱层析结合紫外吸收观察的方法分离抗补体组份;采用比浊法观察组份对眼镜蛇毒因子引起的血小板变形的影响;采用MTT法观察组份对异种血清对内皮细胞损伤的影响。结果紫外吸收为196 nm的组份具有抗补体作用,麻黄组份能有效抑制眼镜蛇毒因子引起的血小板变形,高、中、低剂量对血小板变形的抑制率分别为49.8%±10.7%、32.8%±4.2%、22.5%±6.2%;麻黄组份能有效减轻异种血清对内皮细胞的损伤,高、中、低剂量对补体致内皮细胞损伤抑制率分别为88.1%±7.1%、79.8%±3.3%、65.0%±4.5%。结论麻黄抗补体组份对补体依赖性损伤有保护作用。Objective To isolate the anti-complement components from ephedra sinica and investigate its protective effect on complement-dependent injury. Methods Anti-complement components were seprerated by using column chro- matography;turbidimetric assay was used to investigate the effect of anti-complement component on CVF-induced platelet metamorphose;MTl" method was used to investigate the effect of anti-complement components on endothelial cells injury caused by rabbit serum. Results Anti-complement components effectively reduced CVF-induced platelet metamorphose and attenuated the endothelial cells injury caused by rabbit serum. Conclusion Anti-complement components may have protective effect on complement-dependent injury.
