摘 要:目的制备稳定的颅脑损伤合并股骨闭合性骨折大鼠模型,研究颅脑损伤后SD大鼠股骨折愈合过程中骨痂降钙素基因相关肽(CGRP)的表达,分析脑外伤对大鼠股骨折愈合速度的影响机制。方法48只雄性Sprague-Dawley(SD)大鼠随机分成二组:颅脑损伤合并股骨折组;单纯股骨折组,分别于造模后7d、14d、28d分批处死动物,截取骨折端长约10mm左右股骨骨痂标本,应用HE染色,免疫组化法,观察CGRP表达的动态变化。结果免疫组化法测定骨折位点骨痂中CGRP平均阳性细胞百分率:脑外伤合并股骨折组在7d、14d与单纯骨折组比较差异有统计学意义。28d二组比较差异无统计学意义。结论脑外伤后合并股骨折,骨折端CGRP在骨折愈合的早中期表达增高,有利于多种细胞趋化、增殖与分化,促进成骨。Objective To investigate the expression of CGRP in callus of rats after brain injury and femoral fracture, to study the mechanism of brain injury on fracture healing and the relationship between CGRP and enhanced osteogenesis after brain injury. Methods Totally 48 male SD rats were randomly divided into two groups which models were established, fracture with brain injury group{n = 24} and femoral fracture group [n = 24). The rats were sacrificed at 7 day, 14 day, 28 day after modeling, and then cut about 10ram femo- ral callus around the fracture site at the same time. The section preparation was made through routine de- hydration, paraffin imbedding, and femoral callus were examined by HE and immunohistochemisty. Results The mean cell positive rates of CGRP by HE and immunohistochemisty: the comparison between femoral fracture group and fracture with brain injury group at 7 day, 14 day had significantly differences; but they were no significantly differences at 28 day. Conclusion The earlier appearance and increase of CGRP ex- pression are higher than those of femoral fracture at the fracture sites based on the mechanism underlying acceleration on osteogenesis, proliferation, differation, thus can helpful for the promotion of bone healing.
