摘  要:目的观察电针丰隆穴对高脂血症大鼠单核趋化因子(MCP-1)、白细胞介素1γ(IL-1γ)含量的影响。方法将40只健康SD大鼠随机分为正常对照组、高脂饲料组、高脂+普通饲料组、高脂饲料治疗组及高脂+普通饲料治疗组,每组8只。正常组喂普通饲料,其余4组采用高脂饮食饲料饲喂法建立高脂血症大鼠模型,造模28 d后,高脂+普通饲料组、高脂+普通饲料治疗组改用普通饲料喂养,其余3组饲喂方法不变,同时高脂饲料治疗组及高脂+普通饲料治疗组电针丰隆穴,疏密波,频率为2/100 Hz,电流强度2 mA,治疗30 min,每日1次,连续治疗28 d。治疗结束后,检测各组大鼠血脂水平,即总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)含量,流式细胞术(FCM)及酶联免疫吸附法(ELASA)检测各组大鼠腹腔巨噬细胞中单核趋化因子(MCP-1)、白细胞介素1γ(IL-1γ)的表达。结果高脂饲料组大鼠血浆TC、LDL-C与正常对照组相比明显上升(P<0.01);高脂+普通饲料组大鼠血浆TC、LDL-C与高脂饲料组相比较明显下降(P<0.01),较正常对照组仍明显升高(P<0.01);电针丰隆穴治疗后,高脂饲料治疗组与高脂饲料组相比较,TC和LDL-C明显降低(P<0.01),高脂+普通饲料治疗组大鼠血浆中TC、LDL-C与高脂+普通饲料组相比较,明显降低(P<0.01)。高脂饲料组大鼠腹腔巨噬细胞、MCP-1及IL-1γ的表达较正常对照组显著升高(P<0.01);高脂+普通饲料组大鼠腹腔巨噬细胞、MCP-1及IL-1γ的表达较高脂饲料组明显下降(P<0.05),但仍较正常组明显升高(P<0.05或P<0.01);高脂+普通饲料治疗组大鼠腹腔巨噬细胞、MCP-1及IL-1γ的表达较高脂饲料治疗组显著下降(P<0.01)。结论电针丰隆穴能够明显下调高脂血症大鼠巨噬细胞、MCP-1、IL-1γ的表达,对高脂血症具有一定的治疗作用。Objective: To explore the effect of electroacupuncture at Fenglong (ST40) acupoint on inflammatory factors MCP-1 and IL-1γ of rats with hyperlipemia(HLP). Methods: 40 health SD rats were randomied into normal control group,high fat feed group,high fat+normal feed group,high fat feed treatment group,and high fat+ normal feed treatment group,with 8 rats in each group. The high fat feed group was eatablished by feeding the animals with high fat forage for 28 days. After modeling,rats of high fat + normal feed group and high fat + normal feed treatment group were fed bassl forage. EA(2 mA,2 Hz/100 Hz) was applied to bilateral Fenglong(ST40) for 30 min,once daily for 28 days. After electro-acupuncture at Fenglong acupoint for 28 days,blood fat level,total cholesterol (TC) ,triglyceride (TG) ,low-density lipoprotein cholesterol (LDL-C) ,and high-density lipoprotein cholesterol (HDL-C) were tested. Flow cytometry (FCM) and enzyme-linked immunosorbent assay (ELASA) were applied to observe changes of the expression of MCP-1 and IL-1γ in macrophages of hyperlipidemic rats. Resuits: The content of TC and LDL-C showed an apparent increase in high fat feed group as compared with normal control group and high fat + normal feed group (P〈 0.01 ). There were significant reductions in the content of TC and LDL-C after the treatment(P〈0.01 ). The expression of MCP-1 and IL-1γ in macrophages of hyperlipidemic rats showed an apparent increase in high fat feed group as compared with normal control group (P〈 0.01) and high fat + normal feed group (P〈0.05) and high fat + normal feed group still showed an apparent increase compared with normal control group (P〈0.05 or P〈0.01 ). Compared with high fat feed treatment group,the expression of macrophages,MCP-1 and IL-1γ in macrophages of hyperlipidemic rats decreased obviously in high fat + normal feed treatment group. Conclusion: Electro-acupuncture at Fenglong acupoint can not ably decrease
