摘  要:目的探讨白藜芦醇(Res)对大鼠急性脊髓损伤保护作用的剂量效应关系。方法采用Allens脊髓打击损伤模型,将60只SD大鼠随机分为6组,每组10只。A组:假手术组;B组:脊髓打击伤组;C组:50mg/kg Res治疗组;D组:100mg/kg Res治疗组;E组:200mg/kg Res治疗组;F组:300mg/kg Res治疗组。术后24、48、72h运用BBB法分别进行运动功能评分;72h处死动物取材,HE染色、Nissl染色观察脊髓组织病理学变化。结果 BBB评分提示24、48、72h三个时间点C、D、E、F组较B组大鼠后肢运动功能均有不同程度的改善;72h时间点,C、D、E、F组较B组运动功能评分差异有统计学意义(P<0.05);Res组较B组的病理组织结构均有不同程度的改善;E组及F组运动功能评分及病理组织结构优于C组及D组,E组与F组间差异无统计学意义(P>0.05)。结论 Res对大鼠脊髓损伤有较好的保护作用,并呈一定的剂量效应关系;200mg/kg是Res干预SCI的最佳剂量。Objective To investigate the dose-effect relationship of the protective effect of resveratrol (Res) on acute spinal cord injury (SCI) in rats.Methods Rat SCI model was established by Allen's weight hit.A total of 60 rats were randomly assigned into six groups (10 in each):sham group (Group A),control group(Group B),50 mg/kg Res group (Group C),100 mg/kg Res group(Group D),200 mg/kg Res group (Group E) and 300 mg/kg Res group (Group F).Basso Beattle Bresnahan locomotor rating scale (BBB) was used to detect locomotors function at 24 h,48 h and 72 h after operation.Rats were sacrificed at 72 h after operation.HE staining and Nissl staining were used to detect the morphological changes of the spinal cord.Results BBB locomotor rating scale indicated that hindlimb motor function in Groups C,D,E and F was higher than that in Group B at 24 h,48 h and 72 h after operation.There was a significant difference in locomotor rating score between Res injection group and control group (P<0.05).The pathological structure was better in Res injection group than in control group.Motor function score and pathological tissue structure were better in Group E and Group F than in Group C and Group D; there was no significant difference between Group E and Group F (P>0.05).Conclusion Resveratrol has a significant neuroprotective effect on SCI in rats.And there is a dose-effect relationship between the dose of resveratrol injection and spinal cord injury,with 200 mg/kg as the optimal dosage for intervening SCI.
