摘  要:目的:检测神经生长因子(NGF)在子痫前期孕妇血清及胎盘组织中的表达,探讨其与妊娠期高血压疾病的关系。方法:收集42例子痫前期孕妇作为实验组,其中轻度子痫前期(MP)10例,重度子痫前期(SP)32例,将30例正常晚期孕妇作为对照组;采用酶联免疫吸附试验(ELISA)测定产前及产后血清及产后胎盘组织匀浆中NGF浓度,免疫组织化学方法测定胎盘组织中NGF表达。结果:实验组产前血清NGF浓度(ng/L)高于对照组(356.34±81.76 vs 229.23±72.04,P=0.000),SP患者高于MP患者(374.97±81.01 vs 296.73±51.65,P=0.005);实验组产后血清NGF浓度(ng/L)高于对照组(419.78±100.52 vs 279.44±70.15,P=0.000),SP患者高于MP患者(442.38±104.58 vs 347.45±27.08,P=0.007);实验组和对照组产后血清NGF浓度均高于产前(419.78±100.52 vs 356.34±81.76,P=0.002;279.44±70.15 vs 229.23±72.04,P=0.008);实验组胎盘组织匀浆液中NGF浓度高于对照组(166.46±16.91 vs 134.93±17.84,P=0.000),SP患者高于MP患者(170.82±16.71 vs 152.50±7.55,P=0.003);胎盘组织免疫组织化学结果显示,NGF在对照组呈低表达或阴性,MP呈中高表达,SP呈高表达,差异有统计学意义。结论:子痫前期患者血清和胎盘组织中NGF表达增高,重度子痫前期患者NGF增高更明显,提示NGF在子痫前期病情发展过程中可能起到一定作用,其表达水平与病情严重程度有关。Objective : To detect the expression of nerve growth factor (NGF) in serum and placenta of preeclampsia patients, and to reveal the relationship between NGF and hypertensive disorder compli- cating pregnancy (HDCP). Methods: Forty-two cases of preeclampsia served as experimental group including 32 cases with severe preeclampsia (SP) and 10 cases with mild preeclampsia (MP) , and 30 normal pregnant women served as control group. The NGF levels in serum and placental tissue homoge- hate were detected by enzyme-linked immunosorbent assay (ELISA) before and after delivery. The ex- pression of NGF in placenta tissue was detected by immunohistochemistry. Results: The serum NGF levels in the experimental group were significantly higher than those of control group before and after delivery, and those of SP were significantly higher than MP; The serum NGF levels after delivery were notably higher than those before delivery in the two groups ;The NGF levels in placental tissue homoge- hate in the experimental group were significantly higher than those of control group, NGF levels of SPwere significantly higher than those of MP;Immunohistochemistry showed that the control group lowly or negatively expressed NGF, MP patients moderately or highly expressed NGF, SP patients highly ex- pressed NGF, the differences were significant. Conclusion: NGF levels in serum and placenta of pre- eclampsia patients are increased, and the increase is more obvious in SP patient, which suggest that NGF may be related to the development of HDCP. NGF level is correlated with severity of the disease.
