摘  要:为研究紫杉醇对犬乳腺肿瘤细胞(CHMm)周期相关因子bcl-2、p53 mRNA表达的影响,本研究以不同浓度的紫杉醇在不同作用时间处理CHMm,通过SYBR荧光定量PCR检测bcl-2、p53的mRNA表达的变化。结果显示:紫杉醇作用后,CHMm细胞生长受到不同程度抑制,并引起细胞凋亡。在紫杉醇诱导CHMm产生细胞毒性导致细胞死亡率不高的情况下,1μmol/L作用12 h^24 h bcl-2 mRNA降低明显。1μmol/L紫杉醇作用24 h高于0.01μmol/L作用48 h时p53 mRNA升高的比值,差异显著(p<0.01)。表明1μmol/L紫杉醇对抑制原癌基因bcl-2、促进抑癌基因p53的表达效果最好,并具有诱导细胞凋亡的作用。To study the mRNA expressions of bcl-2 and p53 with paclitaxel treated in canine breast tumor cells (CHMm), the CHMm cells were treated with paclitaxel in different concentrations for different time and detected by SYBR Green I based real-time RT-PCR. The results showed that the CHMm growth was inhibited and the rates of apoptosis increased after paclitaxel treatment. In addition, the Bcl-2 mRNA decreased obviously at concentrations of 1 μmol/L for 12 to 24 hours. Compared with 0.01 μmol/L for 48 hours, the p53 mRNA significantly increased at concentrations of 1 μmol/L for 24 hours (p〈0.01). These data suggested that paclitaxel had the effect to inhibit the proto-oncogene bcl-2 and promoted the expression of tumor suppressor gene p53 at concentrations of 1 μmol/L of paclitaxel treatment, which resulted in inducing cell apoptosis to kill the tumor cells.
