摘 要:为给油菜杂交育种和优势组合的选配提供理论依据,利用SSR标记对54份甘蓝型黄籽油菜DNA指纹图谱的构建及遗传多样性进行分析,并采用高通量法快速提取DNA,运用筛选确定的14个引物检测材料的多态性.结果表明:该材料中多态性片段有82条,平均每对引物5.85条,片段在100~900 bp,多态性比率达67.3%.引物的PIC平均值为0.66,说明这些引物总体上具有较好的鉴别能力.利用7对SSR引物能将54份甘蓝型黄籽油菜材料分开,并构建了其指纹图谱,仅SSR4能分辨22个黄籽材料;54份材料的相似系数为0.13~0.67,平均值为0.40.在相似系数为0.45处将54份甘蓝型黄籽油菜分为12类.To provide a theoretical reference for apolegamy of cross breeding and combination of superiority,by using SSR markers,the DNA finger-print of 54 yellow-seeded B.napus was constructed and the genetic diversity was analyzed,DNA was rapidly extracted by high-throughput method,the selected 14 primers were used to determine the polymorphism.Results:82 bands were polymorphic (67.3%),with an average of 5.85 bands per primer,between 100~900 bp fragment,the mean PIC value of these markers was 0.66,indicating a good resolution power.It was available to separate 54 B.napus yellow-seeded materials apart by using seven pairs of SSR primers and their fingerprints were constructed,only SSR4 can distinguish 22 yellow seed materials; the similarity coefficient of the 54 yellow-seeded B.napus was 0.13~0.67,with an average of 0.40.The observed 82 polymorphic markers are sufficient to separate all the 54 materials from 12 categories at the similar coefficient of 0.45.
