摘 要:为了提高弯曲芽孢杆菌降解有机氮的能力,用紫外线照射法诱变弯曲芽孢杆菌,绘制诱变曲线,确定紫外诱变条件,采用纳氏试剂分光光度法测定NH+-N浓度衡量弯曲芽孢杆菌降解有机氮的效果,并对所选育的高产菌株进行遗传稳定性研究.结果表明:最佳紫外线照射时间确定为30 s,菌落的致死率为91.17%,选育出NH+-N高产菌株UV-14-7,NH4+-N浓度达到304.61 mg/L,比出发菌株提高33.60%;菌株UV-14-7经过7次传代,其产生氨氮的浓度在301.01~304.61 mg/L.菌株UV-14-7有良好的遗传稳定性.To improve the organic nitrogen degrading ability of B.flexus by UV mutagenesis,mutagenesis of B.flexus was carried out by UV irradiation,based on which mutagenesis curve was plotted,and the condition for mutagenesis was optimized.The effects of removal of organic nitrogen by B.flexus was measured by the quantity of ammoniacal nitrogen which was produced.The concentration of NH4+-N was determined by Nessler's reagent spectrophotometry,and investigated for genetic stability of the high-producing strain.Results:The optimal time for UV irradiation was 30s,and the total lethality rate of colonies was 91.17%,highly degrading organic nitrogen strain B.flexus UV-14-7 was obtained,and the concentration of NH4+-N reached 304.61 mg/L,which increased by 33.6% as compared with that produced by the original strain.The concentration of NH4+-N produced by strain UV-14-7 after subcultured for seven passages was 301.01~304.61 mg/L,and variance analysis of generation test showed that strain UV-14-7 has good genetic stability.Highly degrading organic nitrogen B.flexus strain UV-14-7 was of stable genetic property.
