摘 要:目的探讨Survivin对慢性缺氧大鼠肺动脉平滑肌细胞电压门控钾通道(Kv)亚型Kv1.5、Kv2.1表达的影响。方法将18只雄性Wistar大鼠随机数字表法分为对照组(C组)、缺氧4周组(H组)、缺氧+给药YM155(小分子Survivin拮抗剂,HY组)组,每组6只。H组及HY组制作缺氧性肺动脉高压模型。采用免疫组织化学法,逆转录-聚合酶链反应和Western Blot方法,观察Kv1.5、Kv2.1在三组大鼠肺动脉平滑肌的表达情况。结果经过4周缺氧后,H组Kv1.5、Kv2.1的mRNA及蛋白质表达明显降低。HY组表达明显增加(P均<0.05),接近于C组(P均>0.05)。在肺内小动脉上,H组Kv1.5、Kv2.1蛋白的表达要明显低于C组(P<0.05),而HY组与C组表达水平接近(P>0.05)。结论 Survivin可以降低慢性缺氧大鼠肺动脉平滑肌细胞的Kv1.5与Kv2.1的表达水平。Objective To investigate the effect of survivin on the gene expressions of voltage-gated potassium channel in chronic hypoxia pulmonary artery smooth muscle of rat. Methods Eighteen age and body weight matched male Wistar rats were randomly divided into control, chronic hypoxic group for 4 weeks and chromc hypoxic group for 4 weeks treated with YM155 (a novel small-molecule survivin suppressant) group. The expression of mRNA and protein of Kvl. 5, Kv2. 1 (the two mainly members of voltage-gated potassium channels family) in pulmonary artery smooth muscle of rats were detected with real time- PCR, immunohistochemical technique and Western Blotting. Results Following 4 weeks exposure to hypoxia, the mRNA and protein expression of Kvl. 5 and Kv2. 1 in pulmonary artery smooth muscle were down-regulated compared with control group (P 〈0.01 ). Contrarily, after treating with YM155, the mRNA expression of Kv1.5 and Kv2. 1 of pulmonary artery smooth muscle of hypoxic rats were up-regulated, and the protein level of Kvl.5 and Kv2. 1 expression of pulmonary artery smooth muscle of by_ poxic rats were both also up-regulated compared with the hypoxic group (P〈0. 01 ). What's more, using immunohistochemical technique, we found that protein expression of Kvl. 5 and Kv2. 1 in distal pulmonary arteries of rats in control group was consistent with the expression in HY group. Conclusion Stttvivin can down-regulate the expression of Kvl. 5 and Kv2.1 in pulmonary artery smooth muscle of hypoxic rats. Voltage-gated potassium channels might be involved in the mechanism of apoptosis mediated by survivin.
