摘 要:目的观察辛伐他汀对慢性心力衰竭(CHF)兔α-辅肌动蛋白2(α-actinin2)表达的影响。方法雄性大耳白兔45只,随机分为正常对照组、CHF模型组和辛伐他汀治疗组。CHF模型组及辛伐他汀治疗组注射盐酸阿霉素2 mg·kg-1,每周1次,连续10周,建立慢性心力衰竭模型;正常对照组给予相同容量的生理盐水。辛伐他汀治疗组在首次注射盐酸阿霉素时给予辛伐他汀,剂量为1.5 mg·kg-1·d-1,持续至第12周;CHF模型组和正常对照组给予相同容积的生理盐水灌胃12周。心脏彩色超声测量兔左心室结构和功能。处死动物留取左心室室壁观察心肌细胞结构的变化,免疫组织化学染色后检测α-actinin2表达水平。结果与正常对照组比较,辛伐他汀治疗组、CHF模型组的左心室收缩末期内径(LVESD)、左心室舒张末期内径(LVEDD)明显增大,左心室射血分数(LVEF)明显下降,差异均有统计学意义(P<0.05);与CHF模型组比较,辛伐他汀治疗组LVESD、LVEDD减小,LVEF升高,差异均有统计学意义(P<0.05)。与正常对照组比较,辛伐他汀治疗组、CHF模型组α-actinin2表达明显减少(P<0.05),辛伐他汀治疗组与CHF模型组相比表达明显增多(P<0.05)。结论辛伐他汀可明显改善CHF兔左心室重塑,提高其心功能,其机制可能与增加α-actinin2表达有关。Objective To investigate the effect of simvastatin on the α-actinin2 of rabbits with chronic heart failure(CHF).Methods Forty-five male rabbits were randomly divided into normal control group,CHF model group and simvastatin treatment group.The rabbits in CHF model group and simvastatin treatment group were injected adriamycin(2 mg·kg^-1)through the auricular vein,once a week for ten weeks to establish the CHF model; the rabbits in normal control group were injected the same volume saline.The simvastatin treatment group was injected simvastatin(1.5 mg·kg^-1·d^-1) at the first time given adriamycin for 12 weeks; CHF model group and normal control group were given the same volume saline for 12 weeks.The left ventricular structure and function were determined by color doppler ultrasonography.Then the rabbits were sacrificed and the left ventricular walls were taken to observe the changes of myocardial cell structures.The immunohistochemical staining were performed to detect the expression of α-actinin2.Results Compared with the normal control group,the left ventricular end-systolic dimension(LVESD),left ventricular end-diastolic dimension(LVEDD) were increased and the left ventricular ejection fraction(LVEF) were decreased in the CHF model group and simvastatin group(P〈 0.05); compared with the CHF model group,the LVESD,LVEDD were decreased and the LVEF was increased in the simvastatin group(P〈 0.05).The expression of α-actinin2 in simvastatin treatment group and CHF model group were significantly lower than those in normal control group(P〈 0.05); the expression of α-actinin2 in simvastain treatment group was significantly higher than that in CHF model group(P〈 0.05).Conclusion Simvastatin can improve cardiac function and left ventricle myocardial remodeling of rabbit with CHF.It may be associated with increased expression of α-actinin2.
