摘 要:目的研究黄连解毒汤对脂多糖(LPS)所致炎症损伤内皮细胞腺苷酸活化蛋白激酶(AMPK)及细胞间黏附分子-1(ICAM-1)的影响。方法培养人脐带静脉内皮细胞,鉴定后加入LPS诱发炎症损伤,将损伤后的内皮细胞分为空白组、对照组(5-氨基-4-咪唑甲酰胺核糖核苷酸干预组)和观察组(黄连解毒汤干预组),用Western blot法检测各组AMPK水平及磷酸化腺苷酸活化蛋白激酶(P-AMPK)的表达,流式细胞仪检测各组ICAM-1表达,噻唑蓝法检测各组细胞的存活情况。结果各实验组间内皮细胞AMPK水平比较差异无统计学意义(P>0.05);与空白组比较,观察组和对照组P-AMPK的表达量显著增高(P<0.05);ICAM-1表达量显著降低(P<0.05),且内皮细胞存活量显著增加(P<0.05)。结论黄连解毒汤可能通过影响AMPK、ICAM-1对炎症损伤内皮细胞起保护作用。Objective To study the effect of huanglianjiedu decoction on adenosine monophosphate-activated protein kinase(AMPK) and intercellular adhesion molecule-1(ICAM-1) in inflammatory endothelial cells induced by lipopolysaccharide(LPS).Methods Human umbilical vein endothelial cells were cultured,and after that LPS was joined into the culture flask of endothelial cells.The endothelial cells after injury were divided into blank group,control group(5-aminoimidazole-4-carboxamide-1-β-D-ribofuranoside intervened) and observation group(Huanglianjiedu decoction intervened).The expression of AMPK and phosphorylation-AMPK(P-AMPK) were measured by Western blot method.The expression of ICAM-1 in each group was measured by flow cytometry and survival rate of cells in each group was measured by methylthiazolyldiphenyl-tetrazolium bromide method.Results The AMPK content of endothelial cells in each group had no significant difference(P〉0.05).Compared with blank group,the P-AMPK expression of the control group and observation group were significantly higher while the expression of ICAM-1 significantly reduced(P〈 0.05),and the amount of survival cell increased significantly(P〈 0.05).Conclusion Huanglianjiedu decoction possibly plays a protective role by affecting the AMPK and ICAM-1 of the inflammatory endothelial cells.
