吴茱萸碱与紫杉醇协同抑制胃癌细胞SGC-7901的研究

摘　　要：采用MTT法检测吴茱萸碱、紫杉醇单独作用和联合作用对胃癌细胞SGC-7901的增殖抑制效果;用流式细胞术评价吴茱萸碱与紫杉醇联合作用于SGC-7901细胞后对其细胞周期分布和诱导凋亡的影响。结果表明:吴茱萸碱与紫杉醇对SGC-7901细胞具有明显的协同抑制作用,且均呈现良好的量效关系;吴茱萸碱明显的降低了紫杉醇的作用浓度,并提升了紫杉醇的G1期周期阻滞和细胞凋亡效果;Western-blot结果表明,二者联合激活了Casapse 3、8、9,进而诱导细胞凋亡。In vitro,the inhibitory effects of evodiamine,paclitaxel either alone or in combination on SGC- 7901 cells were measured by MTT assay.The effects of evodiamine /paclitaxel combination on cell cycle distribution and apoptosis of SGC- 7901 were determined by FACS can cytometer.MTT assay showed that evodiamine /paclitaxel combination could synergistically inhibit the proliferation of SGC- 7901 cells in a dose-dependent manner.Furthermore,the combination of evodiamine /paclitaxel could decrease the action concentration and result in an increase in G1 cell cycle arrest.Finally,the combination of evodiamine and paclitaxel activated the procaspase 3 /8 /9 and induced the apoptosis in SGC- 7901 cells.
