摘  要:采用免疫组化和荧光定量PCR方法,研究了鹅胚胎期皮肤中Wnt10b基因的定位表达及其mRNA表达规律。结果表明:Wnt10b基因最初在鹅背部皮肤毛囊的毛基板上表达,随后在羽芽、缘板和羽嵴中均有表达,在初级毛囊和次级毛囊发育过程中均有表达。Wnt10b基因在整个胚胎期的毛囊发育中一直动态表达,胚胎期第17天和第19天分别出现2个表达高峰期,此时为次级毛囊刚开始发育的阶段。推测Wnt10b基因对初级毛囊和次级毛囊的形成及发育可能起着重要的促进作用。In this study,immunohistochemistry and fluorescent quantitative PCR were used to study the localization of gene expression and mRNA expression of Wnt10b gene in goose embryonic skin.The orientated expression showed that Wnt10b gene was initially expressed at hair placode of goose back skin hair follicles,followed by feather buds,edge boards and crest.It was also expressed in the primary and secondary follicles during the development of hair follicles.The quantitative expression showed that Wnt10b gene was dynamically expressed in the entire embryonic development of hair follicles and reached crest-time at day E17 and E19 respectively.Secondary follicles began to develop at this stage.It is supposed that Wnt10b gene may play an important role in promoting formation and development of primary and secondary follicles.
