VBE-6联合顺铂对人卵巢癌CoC1细胞增殖的抑制作用

摘　　要：目的:探讨黄荆子木脂素VBE-6联合顺铂对体外培养人卵巢癌CoC1细胞增殖的抑制作用。方法:将CoC1细胞培养并分别单独加入不同浓度VBE-6(0.1、1.0、5.0、10.0、20.0μmol/l)、顺铂(10.0μmol/l)单独及联合作用后,用MTT法检测卵巢癌CoC1细胞增殖情况,流式细胞术检测细胞凋亡率和细胞周期分布。结果:MTT法检测结果显示,不同浓度VBE-6组随着作用时间延长及药物浓度增加,CoC1细胞的增殖抑制率呈明显上升趋势,各组间比较,差异有统计学意义(P<0.05);联合用药组明显高于单一用药组,差异有统计学意义(P<0.05)。流式细胞术结果显示,VBE-6能阻滞卵巢癌CoC1细胞于G1期,同时可诱导细胞凋亡发生;随着VBE-6用药浓度增加,细胞凋亡率增高。结论:VBE-6对卵巢癌CoC1细胞具有增殖抑制和诱导凋亡作用,在一定浓度范围内,这种作用呈剂量-时间依赖性;VBE-6与顺铂联合应用具有协同作用。Objective： To explore the inhibiting effect of VBE - 6 combined with cis - platinum on proliferation of human ovarian cancer CoCl cells cultured in vitro. Methods ： CoCl cells were cultured and treated with VBE - 6 of different concentrations （0. 1, 1.0, 5. O, 10. 0, 20. 0 μmol/L） and cis -platinum （10. 0μmol/L） respectively; MTT method was used to detect proliferation of human ovarian cancer CoCl cells; flow cytometry was used to detect cell apoptotic rate and distribution of cell cycle. Results： The results of MTT method showed that inhibition rate of proliferation of human ovarian cancer CoCl cells showed obvious increasing trends with time extension and in- crease of concentration in VBE - 6 groups of different concentrations, there was statistically significant difference among different groups （ P 〈 0. 05 ） ; the inhibition rate in combined administration group was statistically significantly higher than that in single drug therapy group （ P 〈 0. 05 ） . The results of flow cytometry showed that VBE - 6 could arrest human ovarian cancer CoCl ceils at phase G1 and induce cell apoptosis ; cell apoptotic rate increased with the increase of VBE - 6 concentration. Conclusion ： VBE - 6 can inhibit proliferation and in- duce apoptosis of human ovarian cancer CoCl cells, showing dose - dependent and time - dependent manners within a certain concentration range; VBE -6 combined with cis -platinum has synergistic effect.
