摘 要:以人牙胚为材料,通过RNA原位杂交技术检测WNT/β-CATENIN信号通路的关键因子(受体LRP6,转导因子APC和DVL2)在人牙胚的帽状期和钟状期的表达模式.此外,利用Real-time PCR技术验证这些基因在钟状期的门牙和磨牙,以及磨牙的上皮和间充质的mRNA表达水平.结果显示,LRP6、APC和DVL2基因在帽状期和钟状期人牙胚的上皮和间充质均有表达,而且APC和DVL2基因在钟状期的门牙和磨牙的表达存在差异,LRP6和APC在牙胚上皮和间充质的表达存在差异.结果提示,WNT/β-CATENIN信号通路关键因子LRP6、APC和DVL2在人牙齿发育过程中存在重要的调控作用.In this research, expression patterns of WNT/β-CATENIN signaling components, including LRP6, APC and DVL2 were detected in human tooth germ at the cap and bell stages by in situ hybridization. Furthermore the mRNA expression of these genes in incisors and molars at the bell stage, then in epithelium and mesenchyme of molars were also examined by Real-time PCR. In the developing human incisor and molar germs, LRP6, APC and DVL2 were investigated in dental epithelium and mesenchyme at cap and bell stages. There is significant changes in the expression levels of APC and DVL2 of molar when compared with incisor, and there is significant changes in LRP6 and APC of mesenchyme when compared with epithelium. Our results reveal the status of active LRP6, APC and DVL2 signalings in the human tooth germ.
