摘  要:【目的】研究小粒野生稻(Acc.No.101133)褐飞虱抗性QTL和连锁的分子标记,为水稻褐飞虱抗性育种提供理论依据和育种材料。【方法】采用标准苗期集团法对一套小粒野生稻的基因渗入系进行褐飞虱抗虫鉴定,应用Windows QTL Cartographer 2.5软件的复合区间作图法分析小粒野生稻含有的褐飞虱抗性QTL。【结果】IR24的死苗率为92.2%,表现高感;小粒野生稻的死苗率为11.1%,表现高抗;131份基因渗入系的死苗率差异较大,变异范围为15.5%-91.1%,呈偏态型连续分布。鉴定获得3个褐飞虱抗性QTL,来自小粒野生稻的等位基因均能显著降低幼苗死亡率,联合贡献率为58.8%;qBph3位于第3染色体RM570-RM85区间,qBph4位于第4染色体RM335-RM518区间,qBph12位于第12染色体RM309~RMl7区间,以qBph4的效应值最大,效果最稳定。与IR24相比,携带纯合等位基因qBph4的9个基因渗入系的幼苗平均死亡率极显著下降37.7%。【结论】从小粒野生稻基因渗入系中鉴定出3个褐飞虱抗性QTL位点;抗褐飞虱主效QTLqBph4的验证不仅提供了一个有用的QTL,还为水稻的遗传改良提供了一套褐飞虱抗性育种材料。[Objective]Quantitative trait loci(QTL) for brown planthopper (BPH) resistance were identified from the wild rice species Oryza minuta (Ace. No. 101133) in the present study, and molecular mapping of the BPH resistance gene was carried out in order to provide basis and materials on rice BPH resistance breeding. [Method]The BPH resistance of a set of introgression lines derive from Oryza minuta was identified by using standard seedling stage group method. The seedling mortality of introgression line plants with BPH infestation was measured to evaluate the resistance. The QTL for BPH resistance of 131 introgression lines derived from a cross between O. sativa and O. minuta were determined by using composite interval mapping analysis in software Windows QTL Cartographer 2.5. [Result]The seedlingmortality of IR24 and Oryza minuta was 92.2% and 11.1%, which showed a high susceptibility and high resistance to BPH, respectively. The seedling mortality of 131 introgression lines ranged from 15.5% to 91.1%, which presented skewness continuous dis- tribution. Three QTLs for BPH resistance were detected, among them, qBph3 was located in the interval RM570-RM85 on the long arm of chromosome 3, qBph4 in RM335-RM518 on the short arm of chromosomes 4, and qBph12 in RM309- RM17 on the long arm of chromosomes 12; out of which, qBph4 showed the most stable effect being with highest effective value. The alleles derived from wild rice had the effect on decreasing seedling mortality, and their jointing contribution rate was 58.8%. Duplicate lines of 13 introgression lines carrying alleles of O. minuta at qBph4 were tested with BPH infestation. As compared with IR24, the average seedling mortality of all the 9 lines carrying homozygous alleles of O. minuta at qBph4 was significantly decreased by 37.7% (P〈0.01). [Conclusion]Three QTLs for brown planthopper resistance of introgression lines derived from Oryza minuta was mapped.
