摘 要:目的观察调节性T细胞(regulatory T cells,Treg cells)在子宫内膜异位症(endometriosis,EMT)患者外周血中的频率及转录因子Foxp3在异位内膜、在位内膜和正常内膜组织中的表达,旨在探讨Treg细胞及Foxp3与EMT的关系。方法选取35例EMT患者作为研究组,15例因畸胎瘤行手术的患者为对照组,采集两组患者的外周血,应用流式细胞技术检测两组外周血中Treg细胞的频率;留取35例EMT患者的异位内膜组织,20例EMT患者中因不孕或月经异常行宫腔镜检查术者的在位内膜组织,15例因妇科良性疾病行宫腔镜检查术者的正常内膜组织,采用免疫组化法检测比较异位内膜、在位内膜及正常内膜组织中Foxp3的表达。结果 Treg细胞在EMT患者外周血中的频率显著低于对照组(P<0.05);转录因子Foxp3在异位内膜组织中表达的阳性率显著高于在位内膜及正常内膜组织(P<0.05)。结论 Treg细胞频率在EMT患者外周血中显著下降,在异位内膜组织中显著升高,Treg细胞分布失衡与EMT的发病关系密切。Objective To observe the frequency of regulatory T(Treg)cells in peripheral blood of patients with endometriosis(EMT)and the expression of transcription factor Foxp3 in ectopic endometrium,eutopic endometrium and normal endometrium tissues,to explore the relationship between Treg cells,Foxp3 and EMT. Methods 35 EMT cases and 15 cases with teratoma were selected as study group and control group separately,the peripheral blood of two groups were collected,the frequency of Treg cells in peripheral blood were detected by flow cytometry technique;Ectopic endometrium tissues of 35 ETM cases were extracted,eutopic endometrium of 20 patients conducted hysteroscopy examination for infertility or menstrual abnormalities were extracted,normal endometrium tissues of 15 cases conducted hysteroscopy examination for benign gynecologic disease were extracted. The expression of Foxp3 in 3 kind of tissues were detected by immunohistochemical method. Results The frequency of Treg cells in peripheral blood of ETM patients is significantly lower than control group(P〈 0. 05). The positive expression ratio of Foxp3 in ectopic endometrium tissues were significantly higher than that in eutopic endometrium tissues and normal endometrium tissues(P〈 0. 05). Conclusion There is an significant decrease of Treg cells in peripheral blood and an significant increase of Foxp3 in ectopic endometrium tissues of EMT patients,the distribution imbalance of Treg cells was closely associated with the onset of EMT.
