金龙胶囊对荷人肺腺癌A549裸鼠皮下移植瘤生长的影响

摘　　要：目的探讨金龙胶囊对肺腺癌A549移植瘤生长的抑制作用。方法建立裸鼠移植瘤模型,随机分成阴性对照组(A组)、单药金龙胶囊组(B组)、单药顺铂组(C组)和联合用药组(D组)4组,连续用药4周。观察肿瘤生长情况,计算抑瘤率;免疫组化法检测移植瘤微血管密度(microvessel density,MVD);RT-PCR分析各组治疗后VEGF及凋亡基因Bcl-2mRNA表达情况。结果与阴性对照组比较,金龙胶囊治疗组(B、D组)抑瘤率分别为26.94%、51.61%,联合用药组(D组)效果最佳。治疗组微血管密度分别为18.83±0.75、14.17±0.75,联合用药组最为显著,而单药顺铂组与对照组在抑瘤率和微血管密度表达上均无差异(P>0.05)。金龙胶囊治疗组(B、D组)Bcl-2mRNA表达较对照组有显著差异(P<0.001),但两组之间无差异(P=0.369);四组之间在VEGFmRNA表达上无明显差异(P>0.05)。结论金龙胶囊对荷A549裸鼠移植瘤有抑制作用,联合顺铂用药有显著协同作用,其作用机制可能与抑制肿瘤微血管形成、诱导细胞凋亡增加有关。Objective To explore the effect of Jin Long Capsule with or without cisplatin （ DDP） on tumor growth of lung adenocarcinoma A549 cell xenografts in nude mice .Methods After a model of human lung cancer established by subcutaneous transplantation of A 549 cells in nude mice, 24 mice were randomly divided into control group （A group）, Jin Long Capsule group（B group）, DDP group（C group） and combination treatment group（D group）.Treatment continued for 4 weeks.Inhibition rates were evaluated, tumor micro-vessel density（MVD） were determined by immune-histochemistry, the mRNA expression of VEGF and apoptosis-associated gene（Bcl-2） were assessed by RT-PCR analyses.Results The tumor growth inhibition rates of Jin Long Capsule treatment groups （ B、D groups ） were 26.94% and 51.61%respectively, and more effect in combination group .The expression of MVD in two groups above-mentioned were 18.83 ＆#177;0.75 and 14.17 ＆#177; 0.75 respectively.No statistically difference were seen between DDP group and control group .Compared to control group, RT-PCR analyses revealed a decrease in expression of Bcl-2 genes in A549 tumors by Jin Long Capsule treatment groups （ B、D groups ） , and no statistically difference in these two groups .Either group showed no statistically difference in VEGF mRNA expression .Conclusion Jin Long Capsule has inhibitory effect , and has synergetic effect with traditional chemotherapy on tumor growth of lung adencarcinoma A 549 cell xenografts in nude mice through inhibiting angiogenesis , and may enhance the sensitivity of A 549 cells to DDP by inducing cell apoptosis .
