摘 要:以高山红景天种子诱导的愈伤组织为材料,改变MS培养基中氮源和碳源的种类和比例,研究愈伤组织的生长和分化情况,结果表明:MS培养基中硝酸铵含量低而硝酸钾含量相对高时,愈伤组织生长量比对照高,硝酸铵含量高而硝酸钾含量低时,愈伤组织生长量比对照低,蔗糖浓度50g/L时愈伤组织生长速度最快;MS培养基中减少硝酸铵含量而增加硝酸钾含量或保持对照含量时,愈伤组织分化率提高幅度大,减少硝酸钾含量而增加硝酸铵含量或保持对照含量时,愈伤组织分化率提高幅度小,在蔗糖浓度为10、30、50和70g/L时,浓度越低愈伤组织分化率越高。In this paper, the callus induced from Rhodiola sachalinensis seeds as material, effects of changing the types and proportions of nitrogen and carbon sources in MS medium on the callus growth and differentiation were studied. The results showed that when the am- monium nitrate level was low and the potassium nitrate was relatively high in MS medium, the callus growth was higher than control. In contrast, when the ammonium nitrate level was high and the potassium nitrate was low in MS medium, the callus growth was lower than control. When the concentration of sucrose was 50g/L, the callus growth was fastest. Re- ducing the ammonium nitrate level and increasing or maintaining the potassium nitrate levels in the MS medium, the rate of Callus differentiation was increased considerably, whereas reducing the potassium nitrate level and increasing or maintaining the levels of ammonium nitrate, the increasing amplitude of callus differentiation rate was small. The lower in the sucrose concentration, the higher in the callus differentiation rate, when sucrose concentrations were 10, 30, 50 and 70 g/L, respectively.
