摘 要:目的:观察比较四君子汤、右归饮对衰老模型大鼠脑海马线粒体DNA的影响,从基因水平揭示四君子汤延缓衰老的机制。方法:将60只健康雄性Ds大鼠,随机分为5组,除正常空白对照组外,余4组大鼠腹腔注射D-半乳糖致衰老动物模型,然后分别以四君子汤、右归饮、维生素E灌胃,观察其对衰老模型大鼠脑mtDNA易缺失区的影响。结果:四君子汤、维生素E有防御模型大鼠脑mtDNA缺失突变的作用,而右归饮则没有此作用。结论:四君子汤有防御衰老模型大鼠脑线粒体DNA缺失突变的作用,从而提示四君子汤的抗衰老作用机制可能与能改善脑线粒体功能,调节能量代谢的作用有关。Objective:To explore the effect of Sijunzi decoction on mitochondrial DNA in aging rat model and explain the mechanism of delaying aging by Sijunzi decoction on the gene level. Methods:60 healthy male SD rats were randomly divided into five groups. The aging rat model was induced by intraperitoneal injection of D-galactose except for the normal control group and fed with Sijunzi decoction, Yougui decoction and Vitamin E by gavages respectively. The effect of mitoehondrial DNA deletion in rat brain tissue was observed. Results: Sijunzi decoction and Vitamin E could prevent from mitochondrial DNA deletion, while Youguiyin could not. Conclusion:Sijunzi decoction could prevent from the deletion of the brain mitoehondrial DNA, the antiaging mechanism of Sijunzidecoction probably can improve the function of mito chondrial DAN in brain tissue and adjust energy metabolism.
