783只SD大鼠自发性肿瘤的分析研究

摘　　要：目的总结SD大鼠的自发性肿瘤的发生情况,为检测农药、食品、药品等化学品的致癌性及探讨其作用机制提供基础数据。方法将本实验室最近几年来完成的6个致癌试验的800只对照组大鼠(雌雄各半)的肿瘤发生情况进行汇总并分析。结果实际检测783只大鼠(雌性392只,雄性391只),其中患瘤动物504只(雌性331只,雄性173只),共检出1058例肿瘤,肿瘤总发生率为64.37%(雌性84.44%,雄性44.25%),大鼠平均肿瘤数为2.09例/只(雌性2.46,雄性1.4);大鼠单发和多发肿瘤发生率分别为30.90%和33.46%(雌性分别为30.35%和54.08%,雄性分别为31.45%和12.79%),患瘤动物最高肿瘤数为9例。在所检出的肿瘤大多为良性肿瘤(87.01%),以乳腺纤维腺瘤(31.67%)和垂体腺瘤(27.84%)最为常见;肿瘤多发生于乳腺、垂体、软组织、皮肤等脏器或组织。结论本实验室选用的SD大鼠自发性肿瘤发生率较高,且多为良性肿瘤,以乳腺纤维腺瘤和垂体腺瘤最为多见。Objective To make a summary of the spontaneous neoplastic changes in Sprague Dawley（ SD） rat,and provide basic information to evaluate the carcinogenicity and explore the mechanism of the chemicals,such as pesticide,food,drug and so on.Methods Historical data are presented for spontaneous neoplasms from 800 SD rats（ 400 males and 400 females） that served as controls in 6carcinogenicity（ 2 year） studies conducted recently.Results All rats（ 783 total 392 females and 391 males） were valid and there were total1,058 tumor cases in 504 rats（ 331 females and 173 males）.The total tumor incidences were 64.37%（ female 84.44%,male 44.25%,respectively） and the average number of tumor per bearing tumor rat was 2.09（ female 2.46,male 1.4）.The maximum value of rat with tumor was 9.The incidence rate of single tumor and multiple tumor were 30.90% and 33.46%,87.01% of spontaneous neoplasms were benign tumors,and the most common neoplasmas were mammary fibroadenom（ 31.67%） and pituitary adenoma（ 27.84%） in both sexes and the most common organs or tissue where the tumor occurred were mammary,pituitary,soft tissue and skin.Conclusion SD rats have a high incidence rate of spontaneous neoplasms and majority of these cases were benign tumors,in which the most common tumors were mammary fibroadenoma and pituitary adenoma.
