玻连蛋白对肝癌细胞株凋亡的抑制效应及可能的机制

摘　　要：目的了解玻连蛋白(Vitronectin,VTN)对肝肿瘤细胞株SMMC 7721生长的影响及其对抗3,3'-二吲哚甲烷(3,3'-diindolylmethane,DIM)诱导SMMC 7721凋亡的作用,并初步探讨该过程中所涉及的信号分子。方法采用细胞增殖试剂观察VTN对细胞增殖率的影响,采用激光共聚焦显微镜观察VTN微管蛋白形态的影响,采用流式细胞术和Western Blotting观察VTN对抗植物化学物诱导SMMC 7721细胞凋亡及所涉及的信号分子表达变化。结果 VTN可以剂量依赖的方式促进肿瘤细胞的生长,并可抑制细胞凋亡诱导剂对细胞诱导凋亡的作用。结论体外实验条件下,VTN有助于肝癌细胞株SMMC 7721生长并可保护SMMC7721细胞,使其免予受到凋亡诱导剂的作用,提示VTN在肝癌发生过程中可能具有促进肿瘤细胞生长、抑制化学物诱导凋亡的生物学作用。Objective To understand the effects of Vitronectin on cell growth and apoptosis in the cell line of hepatocellular carcinoma,SMMC 7721.Methods The effect of Vitronectin on cell proliferation rate was appraised with the reagent of WST-1; And observe the morphology of β-Tubulin with con-focal microscope.The effect on apoptosis was detected with flow cytometry and the molecular mechanisms were assayed with Western Blotting.Results Vitronectin help to the adherence of cells to Petri dish and could enhance the growth rate of SMMC 7721 with the dose-effect mode.VTN could also inhibit the apoptosis induced by DIM,the mechanism under which might be related to the phospholation of AKT,the expression of survivin,cleaved caspase-9 and cleaved caspase-3.Conclusion Vitronectin might play important roles in the following biological effects,such as sustaining the morphology of the tumor cells,enhancing the adherence as well as anti-apoptosis.
