致瘤农杆菌的鉴定及与转基因741杨基因互作的研究

摘　　要：以从长瘤的转基因741杨和非转基因毛白杨的根际土壤中分离得到致瘤农杆菌为试材,采用切片、染色及活体接种的方法,对其进行了形态和生物学鉴定。结果表明:其形态符合致瘤农杆菌的特征;胡萝卜和番茄苗的致瘤性试验结果为阳性;该菌株在诱导胡萝卜和番茄苗肿瘤发生同时,还发现其有明显地促进杨树组培苗以及番茄实生苗不定根的诱导能力;该菌株回接转基因741杨组培苗后,并未发现与其寄主之间存在基因转移现象。Taking Agrobacterium tumorigenic isolated from the rhizosphere soil of transgenic 741 Populus long tumour and non-GMO popalus tomentosa as experimental materials,with the methods of sections,staining and inoculated respectively,the morphology and biology of two Agrobacterium tumorigenic were identified.The results showed that the morphological characters were accord with the character of the Agrobacterium tumefaciens.Tumorigenicity test of carrot and tomato seedlings was positive;The bacterium induced tumour of carrot and to moto,at the same time,this bacterium could facilitate the rootage and the cancerization of the carrots and the tomato seedlings.The gene flow phenomena could not be found between this bacterium and its host after it was reinoculated to the transgenic 741 Populus.
