摘  要:【目的】观察PM2.5对人外周血淋巴细胞凋亡的影响并探讨其可能机制。【方法】采用0、33、100和300μg/ml PM2.5染毒培养的外周血淋巴细胞24、48 h,分别用流式细胞仪测定淋巴细胞凋亡率,紫外分光光度计测定羟自由基(·OH),高效液相色谱-电化学检测分析法测定8-氧鸟嘌呤(8-oxodG)。【结果】PM2.5染毒细胞24 h时,PM2.5各浓度组(33、100和300μg/ml)淋巴细胞凋亡率与对照组(0μg/ml)比较无显著性差异(P>0.05);100和300μg/ml组淋巴细胞·OH和8-oxodG均稍高于对照组(P<0.05或P<0.01)。PM2.5染毒细胞48 h时,100和300μg/ml组淋巴细胞凋亡率、·OH和8-oxodG均显著高于对照组(P<0.01),且300μg/ml组高于100μg/ml组(P<0.01)。【结论】PM2.5可诱导外周血淋巴细胞凋亡,其机制可能与自由基产生增多并造成DNA氧化损伤有关。[Objective] To observe the effect of PM2.5 on peripheral blood lymphocyte apoptosis and investigate the possible mechanism. [Methods]The peripheral blood lymphocytes were exposed to related PM2.5 at the doses of 0, 33, 100 and 300 μg/ml for 24 hours and 48 hours ~spectively, and lymphocyte apoptotie rate was determined by flow cytometer, hydroxyl free radical (·OH) by ultraviolet spectrophotometer, 8-oxo-2'-deoxyguanosine (8-oxodG) content by high-performance liquid chromatography- electroche- mical detection method. [Results]At 24 h exposure to PM2.5, lymphocyte apoptotic rate in each PM2.5 dose group (33, 100 and 300 μg/ml) had no change (P〉 0.05), while ·OH and 8-oxodG in 100 and 300 μg/ml group slightly increased compared with control group (P 〈 0.05 or P 〈 0.01). At 48 h exposure to PM2.5, lymphocyte apoptotic rate, ·OH and 8-oxodG in 100 and 300 μg/ml group raised significantly compared with control group (P 〈 0.01) and those in 300 μg/ml group were higher than 100μg/ml group (P 〈 0.01). [Conclusion] PM2.5 could induce peripheral blood lymphocyte apoptosis, which was related to oxidative DNA damage induced by ·OH production.
