摘 要:60只SD大鼠随机分为正常对照组、模型对照组、富硒灵芝治疗组。非酒精性脂肪肝病(Non-Alcoholic Fatty Liver Disease,NAFLD)模型建立成功后富硒灵芝治疗组给予富硒灵芝水灌胃治疗,模型对照组给予生理盐水灌胃辅助对比,空白对照组不加以任何干涉,分别于6周和12周处死大鼠各半。大鼠肝脏HE染色显示,治疗6周后富硒灵芝组脂肪变性程度及细胞肿胀相较于模型组明显减轻,炎性细胞浸润及坏死灶较模型对照组明显减少,富硒灵芝12周组大鼠比富硒灵芝6周组肝脏改善效果更佳;治疗6周后,富硒灵芝治疗组与模型对照组相比,血清中乳酸脱氢酶(LDH)、天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、谷胱甘肽(GSH)、丙二醛(MDA),肝脏组织中超氧化物歧化酶(SOD)、MDA和GSH,差异显著(P<0.05);SERBF-1mRNA和蛋白表达:富硒灵芝6周组与模型对照6周组相比,只有微小差异(P>0.05),12周富硒灵芝组与模型对照组相比,差距则具有统计学意义(P<0.05);ACCαmRNA和蛋白表达:富硒灵芝6周组与模型对照6周组相比,差别具有统计学意义(P<0.05);12周时差异更加显著;胰岛素抵抗指数6周时没有改善,12周时呈现出统计学意义(P<0.05)。因而推测,富硒灵芝可通过调控大鼠ACCα的表达、以提高抗氧化能力及改善肝功能来治疗NAFLD,但药效疗程较长,可能需长期服用。60 SD rats were randomly divided into normal control group, model control group, Se-enriched Ganoderma luci- dum treatment group. After non-alcoholic fatty liver disease (NAFLD) model was successfully established, the Se-en- rlched G. lucidurn treatment group received Se-enriched G. lucidum water gavage treatment and the model control group received saline auxiliary for comparison while the control group were treated with no interference. After 6 weeks and 12 weeks, half of the rats were sacrificed each time. HE staining showed that, after 6 weeks' treatment, the steatosis and cell swelling was significantly reduced in the rats with Seenriched G. lucidum treatment. The infiltration and necrosis in inflammatory cell was significantly reduced compared with the model group. The effect of 12 weeks' treatment with Se-enriched G. lucidum was better than that of 6 weeks'. After 6 weeks ' treatment, the level of lactate dehydrogenase (LDH), aspartate aminotransferase (AST), alanine aminotransferase (ALT), serum glutathione glutathione (GSH) ,malondialdehyde (MDA) ,liver superoxide dismutase (SOD) ,liver MDA and liver GSH in the group with Se- enriched G. lucidum treatment changed signiilcantly, compared with the control group, P 〈 0.05. Comparing the SERBF-1 mRNA and protein expression in Se-enriched G. luc/dum 6 weeks group with the expression in control group, there was only minor differences (P 〉 0.05) ,but the difference was statistically significance (P 〈0.05) between 12 weeks group and control group. ACCa mRNA and protein expression:comparing the Se-enriched G. luc/dwn 6 weeks' group with model control group, the difference was statistically significant (P 〈 0.05 ) and the difference was more significant at the 12 weeks' group. The 6 weeks' insulin resistance index (IR), had no improvement, however,it was statistically slgniil- cant (P 〈 0.05) at the 12 weeks' group.
