摘 要:目的观察慢性阻塞性肺疾病(COPD)大鼠模型血浆中白细胞介素(IL)-6、超氧化物歧化酶(SOD)及丙二醛(MDA)表达,以及低分子肝素(LMWH)干预对其表达的影响。方法将30只Wistar大鼠随机均分为COPD模型组、LMWH干预组和正常对照组。正常对照组正常环境下喂养,不予处理;应用熏烟联合气管内注入脂多糖的方法建立COPD模型;LMWH组在建立COPD模型同时皮下注射低分子肝素。观察3组大鼠肺组织病理形态改变,检测大鼠血浆中IL-6、MDA及SOD水平,分析各指标间的相关性。结果模型组及LMWH干预组大鼠血浆IL-6及MDA水平高于正常组,SOD水平低于正常组(F=55.55~117.07,q=5.57~15.29,P〈0.01);干预组大鼠与模型组相比,IL-6及MDA水平明显降低,SOD水平明显升高(q=4.96~7.67,P〈0.01)。3组大鼠的结果综合分析显示,IL-6与SOD水平呈显著负相关(r=-0.791,P〈0.01),IL-6与MDA水平呈显著正相关(r=0.890,P〈0.01)。结论 LMWH在COPD大鼠模型中可能具有抗炎及抗氧化作用,但两种作用之间的关系及具体机制尚待进一步研究。Objective To investigate the expressions of plasma interleukin-6(IL-6),superoxide dismutase(SOD)and malonaldehyde(MDA)in COPD rats,and the effects of low molecular heparin(LMH)on the expressions. Methods Thirty Wistar rats were randomized to COPD model group,LMH-treated group and normal control group.The rats in the normal control group were fed under normal environment;a COPD model was created using combined smoke inhalations and endotracheal instillations of lipopolysaccharide;those in the LMH group were given subcutaneous injection of LMH at the same time of creation of COPD model.The pathology of lung tissues of rats in the three groups were observed,the levels of plasma IL-6,SOD and MDA were detected and the correlation between the parameters was analyzed. Results Compared with the rats in normal group,the levels of IL-6and MDA in model and LMW groups were higher and the SOD were lower(F=55.55-117.07,q=5.57-15.29,P〈0.01),Compared with the model group,the levels of IL-6and MDA in the LMH group were lower and the SOD higher(q=4.96-7.67,P〈0.01).Meta-analysis showed the levels of IL-6were negatively correlated with SOD(r=-0.791,P〈0.01),and IL-6positively correlated with MDA(r=0.890,P〈0.01). Conclusion Low molecular heparin may have actions of anti-inflammatory and anti-oxidant in rat models with chronic obstructive pulmonary disease,but,the relationship between the two actions and their mechanisms need to be further studied.
