复方甘草酸苷对HSP病儿Treg和Th17细胞功能影响

摘　　要：目的探讨复方甘草酸苷对过敏性紫癜(HSP)病儿Treg和Th17细胞功能的影响。方法选取39例HSP病儿,随机分为复方甘草酸苷组20例和糖皮质激素组19例,以22例健康体检儿童作为对照组。治疗前后采集受检者外周静脉血,采用双抗体夹心酶联免疫吸附试验(ELISA)检测外周血清白细胞介素-17(IL-17)、白细胞介素-10(IL-10)和转化生长因子-β(TGF-β)的含量。结果 HSP病儿急性期外周血IL-17水平较对照组显著增高(t=6.58,P<0.01),而血清IL-10、TGF-β水平与对照组比较差异无显著性(P>0.05);复方甘草酸苷组和糖皮质激素组用药后血清IL-17水平较治疗前均显著降低(t=9.27、9.41,P<0.01),前者降低幅度低于糖皮质激素组(t=3.06,P<0.05);糖皮质激素组治疗后血清IL-10、TGF-β水平较治疗前明显降低(t=3.80、3.64,P<0.05),而复方甘草酸苷组治疗前后差异无显著性(P>0.05)。结论复方甘草酸苷可显著降低HSP病儿外周血Th17细胞功能,但对调节性T细胞功能无明显影响。Objective To explore the influence of compound glycyrrhizin（CG）on the functions of Treg and Th17cells in children with Henoch-Schonlein purpura（HSP）. Methods Thirty-nine children with HSP were enrolled and randomized to CG group（20cases）and glucocorticoid（GC）group（19）,and 22healthy children were served as control group.Before and after treatment,peripheral venous blood was collected from the patients,interleukin-17（IL-17）,interleukin-10（IL-10）and transforming growth factor-beta（TGF-β）were detected by enzyme-linked immunosorbent assay（ELISA）. Results In acute phase,the IL-17level in sick children was significantly higher than those in the normal control group（t=6.58,P〈0.01）,but the differences of IL-10and TGF-βwere not significant between the two groups（P〈0.05）.After treatment,serum levels of IL-17in both CG and GC groups were lower than before treatment,the former group being much lower than the GC group（t=3.06,P〈0.05）.In GC group,the levels of IL-10and TGF-βafter therapy were obviously declined versus before（t=3.80,3.64;P〈0.05）,and that were not significantly different in CG group（P〈0.05）. Conclusion Compound glycyrrhizin can significantly reduce the Th17cell function in peripheral blood of children with Henoch-Schonlein purpura,but has no obvious effect on Treg cells.
