岳阳地区血小板捐献者HPA1～17,HLA-A和B基因多态性研究

摘　　要：目的 研究湖南省岳阳地区机采血小板捐献者人类血小板抗原（HPA） 1-17及人类白细胞抗原HLA-A和B基因分布及多态性.方法 分别采用PCR-SSP法和PCR-SSO法对200名岳阳地区机采血小板捐献者进行HPA1-17和HLA-A和B基因分型,计算基因频率、基因型频率,并与其他人群进行比较.结果 岳阳人群HPA1-17系统中,HPA7-14和HPA16-17的基因型均为aa型,不具有多态性;HPA1,2,4和5主要以aa型为主,aa基因型频率分别是0.970 0,0.900 0,0.985 0和0.980 0,未检出bb型;仅在HPA3,6和15中检出bb型,bb基因型频率分别是0.185 0,0.035 0和0.465 0;杂合度最高的是HPA15,其aa,ab和bb基因型频率分别是0.305 0,0.230 0和0.465 0,HPA3杂合度居次,其aa,ab和bb基因型频率分别是0.380 0,0.435 0和0.185 0.HLA-A位点检出12个等位基因,频率最高的3个是A＊11（0.3325）,A＊02（0.295 0）和A＊24（0.205 0）;HLA-B位点检出28个等位基因,频率最高的5个是B＊ 40（60）（0.177 5）,B＊ 46（0.142 5）,B＊ 13（0.100 0）,B＊51（0.082 5）和B＊ 15（75）（0.077 5）.结论 岳阳地区机采血小板捐献者HPA1-17和HLA-A,B基因多态性与南方汉族接近,但也呈现出自身特点,需建立该地HLA/HPA基因分型血小板供者库服务于临床.Objective To investigate the genetic polymorphism of HPA1-17 and HLA-A,B alleles in Yueyang platelet donors.Methods A total of 200 samples from platelet donors of Yueyang were genotyped for HPA1-17 and HLA-A,B locus by PCR-SSP and PCR-SSO and the genetype frequency and gene frequency were calculated.Results The results showed there were no polymorphism in HPA7-14 and HPA16-17,which only expressed-aa type.In HPA-1,2,4 ang 5,the-aa type was more common than-ab type,the-aa type frequencies were 0.970 0,0.900 0,0.985 0 and 0.980 0,respectively.The-bb type was only detected in HPA3,6 and 15,the-bb type frequencies were 0.185 0,0.035 0 and 0.465 0.HPA15 had the greatest heterozygosity with a gene frequency of 0.305 0,0.230 0 and 0.465 0 for HPA15aa,HPA15ab and HPA15bb,respectively.HPA3 showed the second greatest heterozygosity with a gene frequency of 0.380 0,0.435 0 and 0.185 0 for HPA3aa,HPA3ab and HPA3bb,respectively.The number of alleles identified was 12 for HLA-A,28 for HLA-B,in this study.The most frequent HLA-A alleles were A ＊ 11（0.332 5）,A ＊ 02（0.295 0） and A ＊ 24（0.205 0）.The most frequent HLA-B alleles were B ＊ 40（60）（0.177 5）,B＊ 46（0.142 5）,B ＊ 13（0.100 0）,B ＊ 51（0.082 5） and B ＊ 15 （75） （0.077 5）.Conclusion Distribution of the allele polymorphism of HPA1-17 and HLA-A,B in Yueyang platelet donors was similar to southern Chinese Han population,and it exhibited its own characteristic.Should establish a local platelet donor bank of HLA,HPA genetyping for clinical services.
