血清标本保存条件对甲状腺抗体测定的影响

摘　　要：目的 探讨低温长时间保存对血清甲状腺抗体（包括抗甲状腺球蛋白抗体,anti-TgAb和抗甲状腺过氧化酶抗体anti-TPOAb）测定的影响.方法 留取北京协和医院内分泌实验室2013年6月的30份剩余血清,制备甲状腺抗体低、中、高三个浓度的混合血清.低、中、高三个浓度血清分别分装在54支eppendorf管中,盖上盖子,取6支测定甲状腺抗体水平,剩余48支密封于20℃或者80℃保存3个月,观察甲状腺抗体检测结果是否有变化.使用罗氏E170电化学发光分析仪检测原始标本和保存3个月标本甲状腺抗体水平,对20℃或80℃保存前后结果进行对比分析.结果 原始的低、中、高三个浓度的anti-TgAb水平（U/ml）分别为493.6±5.6,1 010.0±I0.4和2 746±54, 20℃保存3个月后,分别为450.6±6.1,931.9±6.9和2 268±74;-80℃保存3个月后,分别为462.9±11.3,988.7±12.7和2 205±69,均与保存前测定结果差异无统计学显著性意义（P＞0.05）.原始的低、中、高三个浓度的anti-TPOAb水平（U/ml）分别为170.2±8.5,289.4±5.3和432.0±8.4, 20℃保存3个月后,分别为142.6±4.2,276.1±4.9和413.2±7.5;-80℃保存3个月后,分别为155.2±1.4,278.6±9.6和384.1±8.4,均与保存前测定结果差异无统计学显著性意义（P＞0.05）.结论 血清低温（20℃或80℃）保存对甲状腺抗体检测无影响.Objective To observe the influence of serum anti-thyroglobulin antibody （anti-TgAb） and anti-thyroperoxidase antibody （anti-TPOAb） results with different methods of serum storage.Methods Collected 30 remaining serum in Jun 2013 in Department of Endocrinology,Peking Union Medical College Hospital.Adopted the three mix-up serum into 54 ep pendorf tubes,took 6 tubes to measure thyroid antibody with Roche E170 electrochemiluminescence analyzer,kept the remaining 48 serum in different condition （20℃ or 80℃）,then tested anti-TgAb and anti-TPOAb and observe the differences.Results The original concentration value （U/ml） of anti-TgAb was 493.6 ± 5.6,1 010.0 ± 10.4 and 2 746 ± 54.After storing in 20 ℃ for 3 months,the concentration of anti-TgAb was 450.6 ± 6.1,931.9 ± 6.9 and 2 268 ± 74,respective ly.After storing in 80℃ for 3 months,the concentration of anti-TgAb was 462.9 ± 11.3,988.7 ± 12.7 and 2 205 ± 69,respectively.The original concentration value （U/ml） of anti-TPOAb was 170.2 ± 8.5,289.4 ± 5.3 and 432.0 ± 8.4,respectively.After storing in 20℃ for 3 months,the concentration of anti-TPOAb was 142.6 ± 4.2,276.1 ± 4.9 and 413.2 ±7.5,respectively.After storing in 80℃ for 3 months,the concentration of anti-TPOAb was 155.2 ± 1.4,278.6 ± 9.6 and 384.1±8.4,respectively.Anti-TgAb and anti-TPOAb results stored in-20℃ or-80℃ for 3 months had no significantly difference with original results in three concentration levels （P＞0.05）.Conclusion Low temperature storage of serum is recommended for analysis of anti-TgAb and anti-TPOAb.
