摘 要:目的:优选羌活-独活合提液中总香豆素的大孔吸附树脂纯化工艺条件。方法:采用UV测定总香豆素含量,检测波长300 nm。通过静态吸附-洗脱试验比较不同型号大孔树脂对总香豆素的分离纯化能力,以总香豆素含量为指标,利用单因素试验考察上样液质量浓度、上样量、洗脱剂体积分数及用量对羌活-独活合提液中总香豆素纯化工艺的影响。结果:D101型树脂对总香豆素具有较好的吸附和解吸附性能,最佳工艺条件为上样液质量浓度1.089 g·L-1,吸附流速1 BV·h-1,上样量2.5 BV,加4.5 BV水洗脱,弃去洗脱液,加80%乙醇2.5 BV洗脱,收集洗脱液;总香豆素吸附率、洗脱率及纯度分别为91.03%,84.80%,63.65%。结论:D101型树脂对羌活-独活合提液中总香豆素具有较好的分离纯化效果,优化的纯化工艺操作简单、稳定可行。Objective:To optimize purification process of total coumarins from combined extract of Notopterygii Rhizoma et Radix and Angelicae Pubescentis Radix by macroporous resin.Method:UV was employed to determine the content of total coumarins with detection wavelength at 300 nm.Types of resin for seperation and purification of total coumarins were screened by static adsorption-elution test,taking the content of total coumarins as index,single factor tests were adopted to optimize purification conditions with the concentration of sample solution,sample volume,eluent concentration and amount as factors.Result:D101 macroporous resin had adsorption and disadsorption ability for total coumarins,optimum purification conditions were as follows:the concentration of sample solution 1.089 g · L-1,adsorption flow rate of 1 BV ·h-1,sample volume of 2.5 BV,eluted with 4.5 BV of water and 2.5 BV of 80% ethanol; adsorption rate,desorption rate and purity of total coumarins were 91.03%,84.80% and 63.65%,respectively.Conclusion:Total coumarins from from combined extract of Notopterygii Rhizoma et Radix and Angelicae Pubescentis Radix could be purified effectively with D101 resin,this optimized process was simple,stable and feasible.
