HPLC测定天山花楸叶提取物中6种黄酮成分

摘　　要：目的：HPLC梯度洗脱法同时测定天山花楸叶提取物中的芦丁、金丝桃苷、异槲皮苷、槲皮素-3-O-（6＂-O-丙二酰基）-β-D-葡萄糖苷、紫云英苷、山柰素-3-O-（6＂-O-丙二酰基）-β-D-葡萄糖苷.方法：采用Waters C18色谱柱（4.6 mm × 150 mm,5 μm）,乙腈-0.4％磷酸溶液为流动相梯度洗脱,运行时间30.0 min,平衡时间10 min,流速1.0 mL· min-,柱温25℃,波长360 nm.结果：芦丁、金丝桃苷、异槲皮苷、槲皮素-3-0-（6＂-0-丙二酰基）-β-D-葡萄糖苷、紫云英苷、山柰素-3-0-（6＂-0-丙二酰基）-β-D-葡萄糖苷分别在3.3 ～39.5,6.2 ～74.1,12.3 ～ 147.5,14.3 ～ 171.2,7.5 ～90.4,15.6～187.5 mg·L-1与峰面积呈良好的线性关系;平均加样回收率分别为98.1％,97.5％,94.5％,95.8％,102.2％,97.9％,其RSD分别为1.37％,1.57％,1.02％,1.77％,2.48％,1.38％.结论：该法操作简便、快捷、准确、稳定性及重复性好,可用于对天山花楸中6个成分的含量测定.Objective：A high-performance liquid chromatographic method was established to evaluate the quality of Sorbus tianschanica with a simultaneous determination of six compounds [rutin,hyperoside,isoquercitrin,quercetin-3-O-（6＂-O-malonyl）-β-D-glucoside,kaempferol-3-O-β-D-glucopyranoside and kaempferol-3-O-（6＂-O-malonyl）-β-D-glucpyranoside].Method：The Diamonsil C18 （4.6 mm × 250 mm,5 μm） column was adopted and diluted with acetonitrile-0.4％ potassium dihydrogen phosphate gradiently at the wavelength of 360 nm.The flow rate was 1.0 mL ·min-1 and the column temperature was 25 ℃.Result：The method had good linearity over the range of 3.3-39.5,6.2-74.1,12.3-147.5,14.3-171.2,7.5-90.4,15.6-187.5 mg ·L-1 for rutin,hyperoside,isoquercitrin,quercetin-3-O-（6＂-O-malonyl）-β-D-glucoside,kaempferol3-O-β-D-glucopyranoside,kaempferol-3-O-（6＂-O-malonyl）-β-D-glucopyranoside,respectively.The average recovery （n=6） of six components were 98.1％,97.5％,94.5％,95.8％,102.2％,and 97.9％,with RSDsof 11.37％,1.57％,1.02％,1.77％,2.48％,and 1.38％ （n =6）.Conclusion：The developed method is convenient,rapid and accurate with high repeatability and stability,it is suitable for the content determination.
