荆花胃康胶丸对小鼠体内H.pylori的根除作用及对胃黏膜上皮细胞形态的影响

摘　　要：[目的]观察荆花胃康胶丸对幽门螺杆菌(H.pylori)感染小鼠的体内杀菌作用及对胃黏膜上皮细胞超微形态的影响。[方法]55只SPF级KM小鼠采用短期免疫抑制联合经口感染方法建立H.pylori感染模型,随机分为空白对照组、模型对照组、荆花胃康组、三联组、联合给药组。荆花胃康组给予荆花胃康挥发油4周;三联组给予兰索拉唑、甲硝唑、克拉霉素1周;联合治疗组给予三联+荆花胃康1周后,单独继续给予荆花胃康3周;模型对照组给予灭菌注射用水;空白对照组不予干预。所有动物均于给药4周后处死,通过快速尿素酶试验、Warthin-Starry染色判断H.pylori根除率,扫描电镜下观察胃黏膜上皮细胞超微形态。[结果]荆花胃康组H.pylori根除率为4/10,三联组7/10,联合给药组8/10,差异无统计学意义;联合给药组胃黏膜上皮细胞形态最优,达到与空白对照组相当的水平,三联组次之,荆花胃康组效应最弱。[结论]荆花胃康胶丸具有小鼠体内根除H.pylori的作用,其与三联疗法联合可有效促进胃黏膜上皮细胞形态结构的恢复。[Objective]To observe bactericidal activity against H.pylori of Jinghua Weikang Capsule and its effect on the ultrastructure of gastric mucosa epithelial cells in vivo.[Methods] Fifty-five SPF KM mice were infected by H.pylori by short-term immunosuppression combined with oral inoculation of H.pylori,which were randomly divided into blank group,control group,Jinghua Weikang group,triple therapy group and combination therapy group.Jinghua Weikang group was administered with Jinghua Weikang volatile oil for 4 weeks,triple therapy group with lansoprazole,metronidazole and clarithromycin for 1 week,combination therapy group with Jinghua Weikang volatile oil and triple drugs for the first week,then only Jinghua Weikang volatile oil for the next 3 weeks,and the control group with sterile water for 4 weeks.All the subjects were sacrificed at the end of the 4th week.The H.pylori eradication rate was judged through rapid urease test and Warthin-Starry staining,and the ultrastructure of gastric mucosa epithelial cells was observed by scanning electron microscope.[Results] The H.pylori eradication rate was 4/10 in Jinghua Weikang group,7/10 in triple therapy group and 8/10 in combination therapy group which had no statistical significance.The optimal result of gastric mucosa epithelial cells ultrastructure was shown in combination therapy group which was equal to the blank group,and the triple therapy group took the second place while the Jinghua Weikang group the third.[Conclusion] Jinghua Weikang Capsule has the bactericidal activity a gainst H.pylori in mice,and combined with triple therapy it can improve the recovery of the structure of gastric mucosa epithelial cells.
