摘 要:目的:观察电针及其预处理对慢性应激抑郁模型(CUMS)大鼠海马中5-羟色胺(5-HT)及5-羟色胺转运体(5-HTT)表达的影响,探讨5-HT、5-HTT在大鼠实验性抑郁症发病过程中的作用机制及电针干预的影响。方法:将60只清洁健康雄性SD大鼠,随机分为空白组、模型组、脉冲电针组、电针预处理组及百忧解组,每组12只。孤养结合CUMS 21天复制抑郁症模型,通过开野试验观察大鼠行为学变化,采用免疫组织化学法检测5-HT、5-HTT在大鼠海马中的表达情况。结果:模型组大鼠开野实验水平运动及垂直运动明显减少,脉冲电针组、电针预处理组及百忧解组均可改善此变化;模型组大鼠海马内5-HT、5-HTT表达均明显降低,脉冲电针组、电针预处理组及百忧解组可逆转此病理变化,在5-HT方面,两组电针治疗及百忧解治疗之间无明显统计学差异;在5-HTT方面,电针预处理有上调趋势,但无统计学意义,较脉冲电针与百忧解组疗效稍逊色。结论:抑郁大鼠的5-HT、5-HTT含量低于正常,电针治疗抑郁症可能通过升高5-HT、5-HTT的含量而发挥抗抑郁作用。电针预处理与脉冲电针治疗抑郁症效果差异不明显。Objective:To investigate the effect on the expression of 5-HT,5-HTT in the hippocampus of rats with chronic unpredictable mild stress by electro -acupuncture ( EA) treatment and EA pretreatment , to eluci-date the possible role of 5-HT,5-HTT in the development of experimental depression and the effect of EA on them.Methods:60 adult male SD rats were randomly divided into normal control group (n=12), model group ( n=12 ) , pulsed EA group ( n=12 ) , EA pretreatment group ( n=12 ) and fluoxetine group ( n=12 ) .Depres-sive model was established by lonely raising and chronic unpredictable mild stress for 21 days.Open-field test was performed to test the behavior of rats .The expressions of hippocampus 5-HT,5-HTT were detected by immunohistochemistry technique .Results:The cross and rear scores of the model group were significantly lower than those in normal control group , while pulsed EA group , EA pretreatment group and fluoxetine group could reverse it.The expressions of hippocampus 5-HT,5-HTT in model group were decreased significantly .The three kinds of treatments could improve these pathological change , and there was no statistical difference among these three treatments in the aspect of increasing 5-HT.While in the aspect of increasing 5-HTT, EA pre-treatment had the tendency of increasing 5-HTT,and the effect of EA and fluoxetine were better than the EA pretreatment .Conclusion:The levels of 5-HT,5-HTT in hippocampus of depression rats are lower than nor-mal.EA treatments may have antagonism against depression by increasing the levels of 5-HT,5-HTT in hip-pocampus ,and there is no difference between the effects of pulsed EA treatment and EA pretreatment on depres-sion .
