Effect of Soaking Seeds with Betaine on Seed Germination Characteristics of White Clover(Trifolium repens L.) under the Simulated Drought Stress

摘  要:以‘海发’和‘拉丁诺’白三叶种子为试材,采用15%和20%PEG6000渗透液模拟干旱胁迫,研究不同程度干旱对白三叶种子萌发特性的影响及不同浓度(0、100mg/L、200mg/L、300mg/L、400mg/L、500mg/L及600mg/L)甜菜碱浸种提高干旱胁迫下白三叶种子萌发能力的效应,为深入了解白三叶种子耐旱性和甜菜碱的作用效果提供理论依据。15%PEG胁迫显著降低了‘拉丁诺’种子的发芽率和发芽势,而对‘海发’无影响,直到20%PEG时‘海发’的发芽势才明显下降;‘海发’的发芽指数在15%和20%PEG时都显著高于‘拉丁诺’,而‘拉丁诺’的活力指数、胚根长、单株鲜重受抑制程度比‘海发’小,单株干重增加比‘海发’大。在干旱胁迫下,‘海发’种子发芽潜力和速率好于‘拉丁诺’,而‘拉丁诺’种子幼苗适应环境的能力大于‘海发’。400mg/L浓度的甜菜碱浸种对提高正常水分下和干旱胁迫下2个白三叶品种种子的萌发能力效果最佳,可有效提高幼苗适应环境的能力。'Haifa' and 'Ladino' white clover cultivars were used to study the effects of different drought stress on seed germination characteristics and the improvement of seed germination ability with soaking seeds with different betaine concentrations (0, 100, 200, 300, 400, 500 and 600mg/L) under 15 and 20 PEG 6000 osmotic stress for further understanding of white clover seed drought tolerance and the effects of betaine. 15% PEG significantly reduced the seed germination rate and germination energy of 'Ladino', but no effect on 'Haifa'. Under 20' PEG, the germination rate and germination energy of 'Haifa' was significantly decreased. The germination index of 'Haifa' were significantly higher than that of 'Ladino' under both 15' PEG and 20' PEG, however, the degrees of inhibition on vigor index, radi- cle length and fresh weight of 'Ladino' was less than that of 'Haifa'. 'Ladino' had a higher increase than 'Haifa' in dry weight per plant. In conclusion, 'Haifa' seeds had better germinate speed and germinating ability than 'I.adino;, while seedlings of 'Ladino ' showed a stronger ability to adapt environment than 'Haifa' under drought stress. Soaking seeds with 400mg/L of betaine showed the best effects on impro- ving seed germination ability in two white clovers under normal water and drought stress, and improved the ability of seedlings to adapt to the environment effectively.
