摘  要:目的探讨香椿子正丁醇提取物对脑缺血再灌注介导大鼠多器官功能障碍综合征的保护作用。方法大脑中动脉线栓法制作大鼠局灶性脑缺血再灌注模型。动物随机分为假手术组、缺血模型组、溶媒对照组、香椿子正丁醇提取物(20mg/kg.d,30mg/kg·d)治疗组、阿司匹林阳性对照组。手术后断头取脑,行神经行为学评分判断神经功能损伤程度,称重法测量脑组织含水量。取肝和肺组织,分光光度法测定丙二醛(malondialdehyde,MDA)、一氧化氮(nitrogen monoxidum,NO)含量和超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧物酶(glutathione peroxidase,GSH-Px)和黄嘌呤氧化酶(xanthine oxidase,XO)活力的变化。结果 1香椿子正丁醇提取物可以改善脑缺血再灌注大鼠神经行为学评分(P﹤0.05)、降低脑缺血再灌注大鼠脑组织含水量(P﹤0.05),从而起到神经保护作用;2香椿子正丁醇提取物可减少肝和肺组织MDA和NO的生成(P﹤0.05)、抑制其SOD、GSH-Px、XO活力升高(P﹤0.05)。结论香椿子正丁醇提取物可通过其抗氧化应激效应对脑缺血介导的多器官功能障碍综合征发挥保护作用。Objective To investigate the protective effects of n - Butanol extract of Chinese toon seeds on multiple organ dysfunc- tion syndrome caused by brain ischemia - reperfusion in rats in vivo. Methods Male SD rats were divided into 6 groups ( sham - operated, isehemie, vehicle and treated group (lower dose and higher dose), positive control group). Before ischemia -reperfu- sion impairment, n - Butanol extract of Chinese toon seeds (20mg/kg. d, 30mg/kg. d) was administrated by intragastrie adminis- tration for 7 days in treated group. After consciousness, neurology deficit score was evaluated. The water extent of brain tissue was determined by weighing method. Levels of malondialdehyde ( MDA ) and nitrogen monoxide ( NO ) , activity of superoxide dis- mutase (SOD), glutathione peroxidase (GSH -Px) and xanthine oxidase (XO) in liver and lung tissue were detected by spec- trophotometer. Results n - Butanol extract of Chinese toon seeds improved the neurology deficit score and reduced the water con- tent of brain ( P 〈 O. 05 ) ; An elevation of SOD, GSH - Px and XO activity and content of MDA and NO in liver and lung tissue after brain ischemia could be inhibited by n - Butanol extract of Chinese toon seeds ( P 〈 0.05 ). Conclusion n - Butanol extract of Chinese toon seeds alleviates multiple organ dysfunction syndrome caused by brain ischemia - reperfusion in rats, which are partly mediated by its anti -oxidative stress effect.
