摘  要:目的基于药效学试验来优选并建立土茯苓(断面红棕色及类白色)药材中总鞣质的最佳提取工艺。方法采用单因素试验、L9(34)正交优化试验及紫外-可见分光光度法,研究提取溶剂浓度、提取时间及次数、料液比对土茯苓(断面红棕色及类白色)药材中总鞣质含量的影响。结果以总鞣质为指标,断面红棕色土茯苓药材最佳提取工艺为:70%丙酮,超声提取40 min,料液比为1.5 g∶25 ml,提取3次;而断面类白色土茯苓药材则为:60%丙酮,超声提取40 min,料液比为1.5 g∶25 ml,提取2次。结论通过正交试验优选的提取工艺方法可行、重复性好、总鞣质提取率高,对土茯苓(断面红棕色及类白色)药材中筛选关键有效部位的药效学试验提供了最佳给药组。Objective Based on the pharmacodynamic test, to optimize and establish the extraction condition of total tannin in Smilax glabra with reddish brown and off - white colored by orthogonal experiment. Methods According to the single factor experi- ment,L9 (34) orthogonal experiment and UV- Vis spectrophotometry,studied the effect of concentration of extraction solvent,ex- traction time,extraction times and solid -liquid ratio on Smilax glabra in total tannin extraction rate. Results The total tannin were investigated, optimum extraction condition of Smilax glabra with reddish brown has been established as follows : the concentration of acetone is 70% , the ultrasonic extraction time is 40 minutes, the ratio of material to liquid is 1.5 g: 25 ml and extraction 3 times; optimum extraction condition of Smilax glabra with off - white colored has been established as follows : the concentration of acetone is 60% ,the ultrasonic extraction time is 40 minutes, the ratio of material to liquid is 1.5 g: 25 ml and extraction 2 times. Conclu- sion Through orthogonal test method is feasible extraction process economic saving high and recovery rate.
