针灸对AD模型大鼠海马轴突生长抑制因子MAG及OMgp表达的影响

摘　　要：目的研究"益肾调督"针灸法对AD模型大鼠海马神经元轴突生长抑制因子MAG及OMgp表达的影响。方法将Wistar大鼠按随机数字表分为4组,即正常组、假手术组、模型组和针灸治疗组,采用双侧海马注射Aβ1-42的模型复制方法,针灸治疗组采用先针刺再艾灸"百会"和"肾俞"穴,通过免疫组化法观察大鼠海马轴突生长抑制因子MAG及OMgp的表达水平。结果模型组大鼠海马轴突生长抑制因子MAG及OMgp阳性细胞平均光密度及平均灰度值与正常组相比显著升高(P<0.01)。针灸治疗组大鼠海马轴突生长抑制因子MAG及OMgp阳性细胞平均光密度及平均灰度值与模型组相比明显降低(P<0.01)。结论针灸可能通过抑制MAG及OMgp的表达,减少轴突损伤,促进轴突再生,进而起到防治AD的作用。Objective To observe the effects of Yishentiaodu acupuncture -moxibustion on the AD （Alzheimers disease） rats with MAG and OMgp of hippocampus axon growth inhibitors, and to investigate the specific mechanism of preventive - treatment on AD by acupuncture - moxibustion. Methods Wistar rats were randomly divided into 4 groups ： the normal group, sham - operated group, model group,and acupuncture- moxibustion group. First,bilateral hippocampus were injected A[31 -42 to make rat model of AD. Then, acupuncture - moxibnstion group selected Baihui points and Shenshu points, and treated with acupuncture and moxi- bustion. Last, detect the expression of hippocampus axon growth inhibitors in rats by the method of immunohistochemisty. Re- suits Compared with normal group and sham - operated group MAG and OMgp expression of model hippocampus is lower obvious- ly than that of them（ P 〈 0.01 ）. However, compared with the model group, MAG and OMgp expression of acupuncture - moxibus- tion group hippocampus mitochondria increased significantly（ P 〈 0.01 ）. Conclusion Acupuncture -moxibustion can effectively inhibition of the expression of the MAG and OMgp,induce the axonal injured and promote axon regeneration, which have the effect of prevention and treatment of AD,
