三白草总木脂素提取工艺优选及对肝星状细胞增殖的抑制作用研究

摘　　要：目的优选三白草总木脂素的最佳醇提工艺及其体外抗肝星状细胞增殖作用的研究。方法通过正交试验法对三白草总木脂素的最佳醇提工艺进行优化,采用MTT比色法测定总木脂素对肝星状细胞增殖的抑制作用。结果确定了三白草总木脂素的最佳醇提工艺条件,即将药材加70%乙醇回流提取2次,每次2 h,每次加醇量为药材量的8倍。总木脂素含量达到57.3%、45.1%及10.6%时,对肝星状细胞增殖抑制率分别为65.2%、50.5%及22.3%。结论三白草总木脂素醇提工艺合理可行,制得的产品质量稳定;总木脂素对肝星状细胞增殖有明显抑制作用。Objective To select the optimum alcohol extraction conditions for total lignans from Saururus chinensis and its inhibition effect on the proliferation of hepatic stellate cells. Methods Orthogonal method was used to optimize alcohol extraction conditions of total lignans and MTT method was employed to investigate the inhibition effect on proliferation of hepatic stellate cells.Results The Optimum extraction technology was as follows: adding 8 times 70% ethanol and reflux extracted 2 hour for 2 times.The inhibition ration reached was 65. 2%,50. 5% and 22. 3%,when the total lignans concentration was 57. 3%,45. 1% and 10.6%. Conclusion The extraction technology of total lignans from Saururus chinensis is reasonable and practical with stable quality,and total lignans had great inhibition effect on the proliferation of hepatic stellate cells.
