摘 要:采用TCA-丙酮沉淀高速离心法,对杉木针叶蛋白质组双向电泳技术中全细胞蛋白的提取、蛋白的裂解、电泳条件等环节进行了优化改良,获得了高重复性、高清晰度的杉木针叶响应低磷胁迫的差异蛋白图谱。在此基础上对低磷处理5 d的杉木进行针叶蛋白提取,并使用ImageMaster 2D Platinum 7.0软件分析比较杉木针叶在对照与低磷处理下的蛋白表达谱。研究表明,图谱中有59个蛋白点存在显著差异,可能与低磷胁迫有关,其中,上调表达点52个,下调表达点7个。In order to obtain the protein profile with high repeatability and resolution under the condition of phosphorus deficiency,the high-speed centrifugation of TCA-acetone precipitation and the optimized electrophoresis including the extraction of whole-cell protein in Chinese fir needles,the protein cleavage and the condition of electrophoresis were used in this study.The protein extraction of fir needles under 5 days low-phosphorus treatment was conducted,and the comparison of protein profile between control and the treatment samples were made by using Image Master 2D Platinum 7 software.The results showed that there were 59 protein points of low phosphorus treatment significantly different from the control sample,which included 52 upregulated and 7 downregulated points.
