摘　　要：目的检测紧密连接蛋白Zo-1、Occludin在间质性膀胱炎(IC)及正常对照组中的表达,研究其表达水平与IC症状评分之间的关系,探讨它们在IC发病中可能扮演的角色。方法采用免疫组化及Western-blot方法检测Zo-1及Occludin在IC组及对照组膀胱组织细胞中的表达,比较两组表达水平的差异;对每个患者分别进行O'Leary-Sant间质性膀胱炎症状评分(ICSI)、问题评分(ICPI)和盆腔症状(PUF)评分,评价Zo-1及Occludin的表达水平与IC症状之间的关系,同时分析ICSI、ICPI、PUF三者之间的相关性。结果在19例对照组膀胱上皮组织的石蜡切片中,可见其阳性染色强,移行上皮层完整,而15例IC患者的膀胱上皮组织阳性染色弱到中等,细胞层变薄或者缺失;IC患者的Zo-1、Occludin的表达均低于对照组,且其表达水平与患者症状评分呈负相关(P值均<0.05);IC患者中ICSI与ICPI、ICPI与PUF、PUF与ICSI总分平均值相关系数分别为:0.801、0.799、0.875(P值均<0.001);结论紧密连接蛋白Zo-1、Occludin在IC中呈低表达,并且与IC症状的严重程度呈负相关;提示Zo-1、Occludin的表达下调可能在间质性膀胱炎的发病中起到一定的作用。Objective To detect the expression of Occludin, Zo-1 in the interstitial cystitis （IC） and control groups, and analyz the relationship between Occludin, Zo-1 and the symptom scores in order to explore their roles in the pathogenesis of IC. Methods Immunohistochemistry and Western-blot were used to detect the expression of Zo-1 and Occludin both in the IC and the normal bladder tissue. And then clarified the relation between Zo-1 and Occludin expression with IC symptoms, as well as the correlation with ICSI, ICPI and PUF. Results 19 cases of bladder epithelial tissue paraffin sections showed strong positive staining and transitional epithelium integrity in the control group, while 15 cases of IC patients with weaker staining and thinner cell layer. The expression of Oceludin, Zo-1 in IC patients was lower than the normal group, and their expression levels were negatively correlated with the severity of the patients＇ symptoms（all P value were 〈0.05）. Furthermore, the correlation coefficients of IC patients ICSI and ICPI, ICPI and PUF, PUF and ICSI were 0.801, 0.799, 0.875 respectively （all Pvalue were 〈0.001）. Conclusions The Occludin, Zo-1 are low -expression in the IC bladder epithelial cells, and their expression levels are negatively correlated with IC symptom scores, which suggests that Zo-1, Occluding may play an important role in the pathogenesis of the IC.
